Interleukin‐8 enhances myocilin expression, Akt‐FoxO3 signaling and myogenic differentiation in rat skeletal muscle cells

Abstract: Interleukin (IL)-8 is released both in visceral adipose tissue and in contracting skeletal muscles. In this study, we examined cellular pathways associated with muscle hypertrophy, chosen on the basis of microRNA profiling, in differentiating rat primary skeletal muscle cells (RSkMC) treated with IL-8 (1 ng/ml) for 11 days. IL-8 increased myocilin expression, Akt phosphorylation, FoxO3 dispersion throughout the cytoplasm, and reduced FoxO3 level. IL-8 decreased the expression of atrogin and MuRF1 and increased… Show more

“…IL-8-neutralizing antibody impairs the differentiation of hMBs ( Polesskaya et al, 2016 ). In contrast, IL-8 treatment decreases the myogenin/MyoD ratio and embryonic MHC expression in rat myoblasts ( Milewska et al, 2019 ). An appropriate level of IL-8 is important for normal myogenesis.…”

“…KEGG pathway enrichment analysis showed that these differentially expressed genes were mainly involved in the PI3K-Akt signaling pathway, regulation of actin cytoskeleton, focal adhesion, and MAPK signaling pathway ( Figure 4F). Among these differentially expressed genes, CCND1 [42], PLCB1 [43], MEF2C [44], and FOXO3 [45,46] have a known function in regulating myogenesis. As a consequence, MEG3 could specifically have important effects on skeletal muscle development.…”
