Interleukin 7 is produced by murine and human keratinocytes.

Abstract: SummaryInterleukin 7 (IL-7) was originally identified as a growth factor for B cell progenitors, and subsequently has been shown to exert proliferative effects on T cell progenitors and mature peripheral T cells as well. Constitutive IL-7 mKNA expression so far had been demonstrated in bone marrow stromal cell lines, thymus, spleen, and among nonlymphoid tissues in liver and kidney. Here we show that both murine and human keratinocytes express IL-7 mRNA and release IL-7 protein in biologically relevant amounts… Show more

“…IL-7 has been detected in human leprosy lesions and was particularly strongly expressed in the tuberculoid form as compared with lesions of the lepromatous form, indicating a beneficial role of IL-7 in cell-mediated immune responses against intracellular pathogens [36]. The cellular source of IL-7 production in such lesions are keratinocytes [25] and/or monocytes [36]. IL-7 is also constitutively secreted by some human B-cell lines, independent of the presence of EBV [37].…”

“…It has recently been demonstrated that IL-7 is produced by epithelial cells (e.g. keratinocytes) [25], and it has been hypothesized that IL-7 production in situ may profoundly influence the quality and magnitude of the local anti-tumour immune response [26,27]. Detection of IL-7 may further aid in characterizing the biological behaviour of colorectal cancer and the interaction between cancer cells and the immune system.…”

“…Interestingly, in vitro IL-7 stimulation was shown to induce CXCR4 expression by naive T cells, suggesting potential homing consequences. 14 IL-7 is constitutively produced by stromal and epithelial cells in the bone marrow, 15 thymus, 16 and lymph nodes 6,[17][18][19] but also in the skin 20 and mucosal tissues. 21 Its affinity for heparan sulfate proteoglycans (HSPGs) on the stromal cells 22,23 suggests that a higher local IL-7 concentration in productive tissues than in plasma may play a role in its functions.…”

“…IL-7 is critical for memory T-cell survival and is principally produced by nonhematopoietic cell types (13)(14)(15)(16). In the spleen and LNs, IL-7 is produced by gp38 + stromal cells known as fibroblastic reticular cells (FRCs) in the T-cell zone, which also produce the chemokines CCL19 and CCL21 to recruit CCR7 + T cells to that site (17).…”
