Interleukin-6/Soluble Interleukin-6 Receptor Signaling Attenuates Proliferation and Invasion, and Induces Morphological Changes of a Newly Established Pleomorphic Malignant Fibrous Histiocytoma Cell Line

Abstract: Pleomorphic malignant fibrous histiocytoma (MFH)is occasionally associated with inflammatory paraneoplastic syndrome (PNS). Recently, we reported that interleukin (IL)-6, one of the candidate cytokines, which induces such systemic inflammatory reaction, may be a tumor-associated factor involved in the pathogenesis and its clinical manifestations of MFH. In the local microenvironment, tumor-induced inflammatory reaction may play a role favoring tumor progression. To clarify the biological relevance of IL-6 in M… Show more

“…There was no significant change in the white blood cell count or the CRP levels prior to and after naproxen treatment. There are several possible mechanisms for the production of inflammatory markers, such as CRP and IL-6 (24)(25)(26). Tumor growth may cause tissue inflammation (24).…”

“…Tumor-associated mononuclear cells may produce IL-6 as an immune cytokine response to tumor growth (25). Cancer cells may increase the production of inflammatory proteins (26). Tumor growth cannot be reduced by naproxen; therefore, radical tumor treatment is required to improve these hematological abnormalities.…”

“…22 In addition, it was recently shown that IL6 plays a pivotal role for proliferation and invasion of malignant fibrous histocytoma (MFH). 23 It is possible that IL6 may have similar functions in MLS/RCLS. The oncogenic potential of IL6 makes it important to determine if this factor is a downstream target of FUS-DDIT3 in MLS.…”

“…Serumfree conditioned medium served as a chemoattractant and was added to the lower chamber. After 16 hours, the migrating cells on the lower side of the filter were fixed with 70% methanol following the protocol previously described (27,28). The chemoinvasion assay was carried out as previously described (29).…”

“…Cell proliferation analysis was done as previously described (28). Briefly, Walker 256 cells (5 Â 10 3 cells) were plated into 24-well plates and cultured in DMEM with 10% FCS for 24 hours.…”
