Interleukin-6 in the Bone Marrow Microenvironment Promotes the Growth and Survival of Neuroblastoma Cells

Abstract: Neuroblastoma, the second most common solid tumor in children, frequently metastasizes to the bone marrow and the bone. Neuroblastoma cells present in the bone marrow stimulate the expression of interleukin-6 (IL-6) by bone marrow stromal cells (BMSC) to activate osteoclasts. Here we have examined whether stromal-derived IL-6 also has a paracrine effect on neuroblastoma cells.

“…IL-6 is a potent activator of osteoclastic activity and its inhibition limits neuroblastoma metastasis in experimental models 170 . This bone marrow-derived stromal IL-6 also directly stimulates the survival and proliferation of IL-6R + neuroblastoma metastases, suggesting a mechanism for the preferred growth of these tumours in the bone 171 .…”

“…The interactions among the multiple factors released from tumor and adjacent microenvironment cells are triggering subsequent responses to influence the behavior of the tumor. 42,43 Unfortunately, it is difficult to devise functional studies to further elucidate the mechanisms and the sequence of molecular changes in a tumor system that includes the tumor microenvironment and specific type of vasculature physiology.…”
