Interleukin-22 attenuates allergic airway inflammation in ovalbumin-induced asthma mouse model

Abstract: Background Allergic asthma is a chronic airway inflammatory disease with a number of cytokines participating in its pathogenesis and progress. Interleukin (IL)-22, which is derived from lymphocytes, acts on epithelial cells and play a role in the chronic airway inflammation. However, the actual role of IL-22 in allergic asthma is still unclear. Therefore, we explored the effect of IL-22 on allergic airway inflammation and airway hyperresponsiveness (AHR) in an ovalbumin (OVA)-induced asthma mou… Show more

“…The structure of the isolated compounds was confirmed by HRESIMS and NMR data analysis. Compared with the literature data, these compounds were identified as pregomisin (3), 24 benzoylgomisin H (5), 25 gomisin O (7), 26 gomisin G (9), 27 (7S,8S,R-biar)-6,6,7,8tetrahydro-12,13-methylenedioxy-1,2,3,14-tetramethoxy-7,8dimethyldibenzo[a,c]cycloocten-9-one (10), 28 gomisin K 1 (11), 29 schisantherin B (12), 30 schisantherin A (13), 30 schisantherin D (15), 30 schsanhenol (17), 27 gomisin L 1 (18), 31 gomisin M 1 (21), 31 gomisin M 2 (22), 31 and schisandrin A (23). 30 Compound 10 was identified in S. chinensis for the first time.…”

“…Then, the tissues were embedded in paraffin wax and sectioned into 4–6 μm for hematoxylin–eosin (H&E) staining. The morphological structure and inflammatory state of the lung tissues were observed using CaseViewer software (3DHISTECH, Hungary) as previously described. , Lung sections were scored on a scale of 0–4 (0, no inflammatory cells; 1, mild inflammatory cells in the bronchial or vascular wall; 2, a ring of inflammatory cells in the bronchial or vascular wall; 3, two–four cell layers of inflammatory cells in the bronchial or vascular wall; 4, severe inflammation with diffuse inflammatory cells, more than four cell layers of inflammatory cells in the bronchial or vascular wall).…”

“…The cell surface-standing IL-22 receptor complex consists of the IL-22Rα and IL-10Rβ receptor chains [ 5 ]. IL-10Rβ is expressed in all cells, including immune cells, whereas IL-22Rα is reportedly expressed in non-hematopoietic cells of the skin, kidney, intestine, liver, and pancreas [ 5 , 10 , 43 ]. To our knowledge, there have been no investigations of IL-22Rα expression in the brain; therefore, our current study examined IL-22 function and IL-22Rα expression in the mouse brain.…”

“…The allergic asthma model was induced by ovalbumin (OVA) according to the previous reports. [18][19][20] Briefly, 10 μg OVA (01641, 97%-100% (HPLC), Sigma, St. Louis, MO, USA), as an allergen, and 2 mg aluminum hydroxide (239186, CAS: 21645-51-2, reagent grade, Sigma), as an adjuvant in 200 µl phosphate buffer saline (PBS, Solarbio, Beijing, China) were intraperitoneally injected into BALB/c nude mice for sensitization on day-0, day-7, and day-14. Subsequently, mice were intranasally exposed to 3.75 μg OVA in 50 μL PBS on day-22, day-24, and day-26.…”
