Interleukin-2-Dependent Allergen-Specific Tissue-Resident Memory Cells Drive Asthma

Abstract: Exposure to inhaled allergens generates T helper 2 (Th2) CD4+ T cells that contribute to episodes of inflammation associated with asthma. Little is known about allergen-specific Th2 memory cells and their contribution to airway inflammation. We generated reagents to understand how endogenous CD4+ T cells specific for a house dust mite (HDM) allergen form and function. After allergen exposure, HDM-specific memory cells persisted as central memory cells in the lymphoid organs and tissue resident memory (Trm) cel… Show more

“…6a–e). Thus, while we see the overall same reduction in T H 2 as Ballesteros-Tato et al , it is equally plausible that low ICOS on the T resident memory cells (T RM ) would mean that both T FH and T RM models (41) could be operable here. The increased T H 1 and T H 17 responses seen in an active EAE model (Fig.…”

“…Resting memory CD4 T cells appear to be largely stable over time by major lineage-defining phenotypic markers, as shown for antigen-specific T FH , T H 1, and T H 2 cells in mouse models (Harrington et al 2008;Hale et al 2013;Hondowicz et al 2016;Tubo et al 2016). Human data support the same conclusion but the antigen-specific data are limited (Locci et al 2013;Bancroft et al 2016;Da Silva Antunes et al 2017).…”

“…In a dust mite model such lung-resident memory cells are sufficient to induce AHR in situ and persist for up to 100 days. 35 In other work, IL-4 receptor (IL-4R) α–responsive T H 2 cells were essential for sustaining AHR but not for inducing acute disease, thereby implying a key role for IL-4/IL-4Rα signaling in perpetuating inflammation in the tissues. 36 …”
