Interleukin 12 gene polymorphisms enhance gastric cancer risk in H pylori infected individuals

Abstract: IL12, formed from p35 and p40 subunits encoded by\ud IL12A and IL12B genes, favours T helper 1 (Th1)\ud differentiation. Th1 lymphocytes prevail over Th2 in\ud H pylori associated chronic gastritis, the first step in\ud H pylori associated gastric carcinogenesis. In this\ud study, 110 patients with non-cardia gastric cancer\ud were compared with 251 patients with benign gastroduodenal\ud diseases to see whether there was any\ud correlation between IL12 gene polymorphisms and\ud H pylori associated gastric aden… Show more

“…In this context it is, however, noteworthy that accumulating evidence would seem to suggest that IL-12 and IL-23 are important mediators in the process that links H pylori infection to gastric cancer. Indeed, polymorphisms of IL-12p40 and p35 genes enhance the risk of non-cardia gastric cancers in H pyloriinfected patients [53] . Moreover, high levels of IL-23 have been documented in human gastric cancers [53] .…”

“…Indeed, polymorphisms of IL-12p40 and p35 genes enhance the risk of non-cardia gastric cancers in H pyloriinfected patients [53] . Moreover, high levels of IL-23 have been documented in human gastric cancers [53] . Nonetheless, no functional study has so far mechanistically linked the activity of IL-12 and IL-23 to gastric cancer.…”

“…IL-12 has previously been implicated in the development of malignant tumors, e.g. Helicobacter pylori -associated gastric cancer [31], HPV-related cervical carcinoma [32], hepatocellular carcinoma [33], and brain tumors [34]. The role of IL-12 in ileal carcinoid development remains to be elucidated, but the data suggest that inflammation and angiogenesis should be evaluated as targets for prevention and/or therapy in ileal carcinoids.…”

“…Because of the functional relevance of IL12, several molecular epidemiologic studies have been done to investigate the association between the IL12A and IL12B polymorphisms and risk of cancers, including nonHodgkin lymphoma (22 -24), hepatocellular carcinoma (25), lung cancer (26), gastric cancer (27,28), and others (29 -32). In the present study, we hypothesized that IL12A and IL12B polymorphisms were associated with cervical cancer risk.…”
