Interleukin-10 Secretion Differentiates Dendritic Cells from Human Liver and Skin

Abstract: Liver dendritic cells (DCs), which may orchestrate the liver's unique immunoregulatory functions, remain poorly characterized. We used a technique of overnight migration from pieces of normal human liver and skin to obtain tissue-derived DCs with minimal culture and no additional cytokine treatment. Liver and skin DCs had a monocyte-like morphology and a partially mature phenotype, expressing myeloid markers, MHCII, and co-stimulatory molecules; but only the skin DCs contained a population of CD1a+ cells. Over… Show more

“…56,57 The little information that is known about human intrahepatic DC function corroborates these general observations. 58,59 Despite the challenges of accessing specimens and extracting cells, a better understanding of how DCs and monocytes function in the intrahepatic environment is important for therapeutic development.…”

“…Briefly, after isolation of lymphocytes, followed by a 13.5% OptiPrep density gradient (Axis Shield, Cambridge, U.K.), CD11c-positive magnetic selection was used to obtain highly purified DCs. Monocyte-derived DCs (MoDCs) from peripheral blood were isolated according to established methods, as previously described (21). Briefly, OptiPrep was added to buffy coats made from whole blood to increase its density to ∼1.1 g/ml, overlayered by two different density solutions of OptiPrep (1.068 and 1.084 g/ml), and centrifuged at 750 3 g for 20 min.…”

“…La sécrétion de chimiokines et de cytokines par les cellules hépatiques a souvent été envisagée comme un des mécanismes favorisant la tolérance hépatique [12,13]. Les cellules dendritiques hépatiques et les cellules NK et NKT ont la capacité de secréter l'IL-10, cytokine immunosuppressive [14,15] …”
