Interlaboratory analysis of teicoplanin plasma concentration assays among Chinese laboratories

Abstract: What is known and objective Teicoplanin is widely used for the treatment of infections caused by drug‐resistant Gram‐positive bacteria. Since there is a good correlation between trough levels and clinical outcome, therapeutic drug monitoring (TDM) is recommended to achieve better clinical curative effects. However, TDM of teicoplanin is not routine in China. So, a programme was initiated in 2017, including both HPLC method establishment and interlaboratory quality assessment, for the measurement of teicoplanin… Show more

“…Moreover, the quantification range was wide with a concentration of up to 500 mg/L. This is beneficial as our clinical target is critically ill patients that have complex clinical characteristics including the prolonged high-dose administration of teicoplanin that depends on perfusion status, infection site, and multidrug-resistance, while most methods using LC-MS/MS or HPLC-UV are available to quantify up to 100 mg/L [14,20,22,23] or 200 mg/L [16].…”

“…The HPLC-UV method using IS for teicoplanin analysis is seldom. One publication reported a practically available HPLC-UV method using piperacillin as the IS [23].…”

“…Remarkably, the increase in formic acid might lead the elution efficiency of RAM and TA3-1 to 80%. Meanwhile, it might cause their degradation and not reach satisfactory recovery (Wang et al, 2020). Thus, a solvent with suitable polarity was explored by adjusting the proportion of methanol, water, and formic acid.…”
