Interferon-γ and B Cell Stimulatory Factor-1 Reciprocally Regulate Ig Isotype Production

Abstract: Gamma interferon (IFN-gamma) and B cell stimulatory factor-1 (BSF-1), also known as interleukin-4, are T cell-derived lymphokines that have potent effects on B cell proliferation and differentiation. They are often secreted by distinct T cell clones. It is now shown that IFN-gamma stimulates the expression of immunoglobulin (Ig) of the IgG2a isotype and inhibits the production of IgG3, IgG1, IgG2b, and IgE. By contrast, BSF-1 has powerful effects in promoting switching to the expression of IgG1 and IgE but mar… Show more

“…As HgCl 2 induces a Th2 response [1,8,9], it is perhaps not surprising that release of Th cells from inhibitory CD8 control might initially result in an exacerbation of Th2 responses (IgE at days 4-7), followed by an increase in Th1 responses (IFNat days 9-17), particularly as this matches the time that Th1 responses have been found to recover in this model [27]. Furthermore, the known inhibitory effect of IFN-on IgE production [28] may explain why the enhancement in IgE was not found after day 7. As HgCl 2 A is / T cell-dependent and Th2-associated, it is possible that an enhancement of regulatory Th1 functions (IFNat days 9-17) might have resulted in the protective effect on the arthritis.…”

“…In addition to the magnitude of humoral immune responses we also analyzed the isotype of anti-Aβ 42 antibodies, since this characteristic has previously been used as an indirect measure of the contribution of Th1 (IgG2a) and Th2 (IgG1) cytokines to the immune response [44][45][46]. Three to six injections with PADRE-Aβ 1-15 -MAP epitope vaccine formulated in Alum generated predominantly IgG1 anti-Aβ antibodies in BALB/c mice (Fig.…”

“…This effect may result from the preferential stimulation of suhsets of helper T lymphocytes whose lymphokines intluence isotype switching in antibody producing cells (6,7), The production of interferon-gamma (7-IFN)or IL-4 by subsets of helper T lymphocytes reciprocally regulates production of lgG2a and IgG 1/E in mice (8). although the situation is less clear in other species.…”
