Interferon alpha induces the expression of retinoblastoma gene product in human Burkitt lymphoma Daudi cells: role in growth regulation.

Abstract: Interferon a (IFN-a) is a regulatory secretory protein with distinctive biological effects such as antiproliferative actions against many tumor cell lines, including human Burkitt lymphoma Daudi cells. The mechanism underlying growth inhibition by IFN-a is not well established. The growth of many mammalian cell types is also regulated by tumor suppressor retinoblastoma (RB) gene product, the RB protein. In the studies presented here, we explored the possible involvement of RB protein in the growth inhibitory ac… Show more

“…This was suggested by a study that looked at involvement of pRb in the growth-regulatory actions of IFN-a by using IFN-a growth-sensitive human Burkitt lymphoma Daudi cells. 39 Treatment of Daudi cells with IFN-a significantly induced pRb protein expression, predominantly as underphosphorylated pRb, by 3-fold to 10-fold, and IFN-a mediated induction of pRb was an early event that preceded the appearance of growth-inhibitory responses and G0/G1 phase arrest of Daudi cells. The mechanism of the observed IFN-a-mediated induction of pRb, however, remains to be delineated.…”

“…Cell Cycle Analysis-Cell cycle distribution was determined by staining with propidium iodide (Sigma) as described (26). Briefly, cells were stained with propidium iodide and passed through the beam of an argon ion laser turned to 514 (FAC/Scan; Becton Dickinson).…”

“…Cell extracts were prepared as described (26). Cell lysate containing equal amounts of total protein (15-25 g) were resolved on a 10% SDSpolyacrylamide gel electrophoresis, followed by probing with an antiBcl-2 mAb (Neomarkers Inc.) using alkaline phosphatase-conjugated second antibody (7,8).…”

“…In vivo, they up-regulate expression of tumor-specific antigens, natural killer, and T cell function (7)(8)(9). IFNs also activate growth suppressive proteins such as pRb (10,11), down-regulate c-Myc (12), and suppress activity of transcription factor E2F (13). Protein kinase R and ribonuclease L, which inhibit viral growth in IFN-treated cells (14,15), also play a role in growth suppression (16,17).…”
