Besides an antigen-presenting function and ability to induce antitumor immune responses, dendritic cells (DCs) possess a direct tumoricidal activity. We previously reported that monocyte-derived IFNα-induced DCs (IFN-DCs) of glioblastoma multiforme patients express low levels of membrane TNFα molecule (mTNFα) and have impaired TNFα/TNF-R1-mediated cytotoxicity against immortalized tumor cell line HEp-2. However, whether the observed defect could affect killer activity of glioma patient DCs against autologous tumor cells remained unclear. Here, we show that donor IFN-DCs possess cytotoxic activity against glioblastoma cell lines derived from a primary tumor culture. Granule-mediated and TNFα/TNF-R1-dependent pathways were established as the main mechanisms underlying cytotoxic activity of IFN-DCs. Glioblastoma patient IFN-DCs showed lower cytotoxicity against autologous glioblastoma cells sensitive to TNFα/TNFR1-mediated lysis, which was associated with low TNFα mRNA expression and high TACE/ADAM-17 enzyme activity. Recombinant IL-2 (rIL-2) and human double-stranded DNA (dsDNA) increased 1.5-fold cytotoxic activity of patient IFN-DCs against autologous glioblastoma cells. dsDNA, but not rIL-2, enhanced the expression of TNFα mRNA and decreased expression and activity of TACE/ADAM-17 enzyme. In addition, dsDNA and rIL-2 stimulated the expression of perforin and granzyme B (in the presence of dsDNA), suggesting the possibility of enhancing DC cytotoxicity against autologous glioblastoma cells via various mechanisms.

Интерфероны I типа являются мощными индукторами дифференцировки моноцитов в дендритные клетки (ДК), однако чувствительность таких ДК к толерогенному эффекту глюкокор-тикоидов ранее не исследовалась. Целью работы явилось изучение влияния дексаметазона на со-зревание и функции интерферон-альфа-индуцированных ДК (IFN-ДК) здоровых доноров. ДК генерировали из моноцитов крови, которые культивировали в течение 5 суток с GM-CSF и IFNα в отсутствие и присутствии декасаметазона (10 -6 M), вносимого на 3 сутки. Добавление дексаметазо-на блокировало созревание IFN-ДК, что проявлялось возрастанием доли CD14 + клеток и снижени-ем содержания CD83 + клеток. Дексаметазон не оказывал значимого влияния на экспрессию HLA-DR, CD86 и B7-H1, однако 2-кратно усиливал экспрессию толерогенной молекулы TLR-2. Наряду с подавлением созревания IFN-ДК, дексаметазон ингибировал продукцию ими провоспалитель-ных/Th1-цитокинов (TNFα, IL-1, IL-2, IFNγ, IL-12) и хемокинов (MIP-1α, RANTES). IFN-ДК, ге-нерированные в присутствии дексаметазона, отличались 2-кратным снижением аллостимуляторной активности в смешанной культуре лимфоцитов (СКЛ). При этом способность IFN-ДК стимулиро-вать пролиферативный ответ Т-клеток в алло-СКЛ прямо коррелирует с экспрессией на ДК моле-кулы CD83 и обратно -с экспрессией СD14 и TLR-2. Оценка Th1-/Th2-поляризующей активности IFN-ДК показала, что дексаметазон оказывал выраженное ингибирующее влияние на способность ДК стимулировать Т-клетки к продукции IFNγ, тогда как супрессорный эффект на способность ДК стимулировать продукцию IL-6 был менее выраженным, что свидетельствует о доминировании Th2-поляризующей активности IFN-ДК под влиянием дексаметазона. В целом показано, что IFN-ДК чувствительны к толерогенному действию дексаметазона и, следовательно, могут опосредовать им-муномодулирующий эффект глюкокортикоидной терапии, а также рассматриваться в качестве но-вых кандидатов для разработки толерогенных лечебных ДК-вакцин при аутоиммунной патологии.

METHODS. To address a potential role for Mtb-infected RPE cells in the development of uveitis, we delineated the response to Mtb infection in human RPE cells and primary human macrophages, the main target cell of Mtb. Primary human RPE cells, the human RPE cell line ARPE-19, and monocyte-derived proinflammatory M1 and anti-inflammatory M2 macrophages were infected with DsRed-expressing Mtb strain H37Rv. Infection rates and clearance were addressed along with RNA sequencing analysis, a confirmation analysis by dual-color reverse-transcriptase multiplex ligation-dependent probe amplification (dcRT-MLPA) and cytokine secretion.RESULTS. RPE cells robustly controlled intracellular outgrowth of Mtb early after infection. The response in RPE cells to control Mtb survival was dominated by interferon (IFN) signaling and further characterized by prominent regulation of cell death/survival-associated genes and low-level production of Th1-associated cytokines. In contrast, macrophages engaged a plethora of responses including IFN signaling and communication between innate and adaptive immune cells to induce granuloma formation. CONCLUSIONS.Together, our data demonstrate that RPE cells display a strong response to Mtb infection that appears, however, incomplete in comparison to the macrophage response to Mtb. The RPE response might reflect a balance between mechanisms aimed at Mtb eradication and mechanisms that limit retinal inflammation.
