Intercellular Adhesion Molecule-5 Induces Dendritic Outgrowth by Homophilic Adhesion

Abstract: Intercellular adhesion molecule-5 (ICAM-5) is a dendritically polarized membrane glycoprotein in telencephalic neurons, which shows heterophilic binding to leukocyte β2-integrins. Here, we show that the human ICAM-5 protein interacts in a homophilic manner through the binding of the immunoglobulin domain 1 to domains 4–5. Surface coated ICAM-5-Fc promoted dendritic outgrowth and arborization of ICAM- 5–expressing hippocampal neurons. During dendritogenesis in developing rat brain, ICAM-5 was in monomer form, w… Show more

“…The expression of ICAM5 is restricted to the mammalian brain telencephalon (24) but there is no homolog in invertebrates and lower vertebrates. The developmental appearance of ICAM5 parallels the time of dendritic elongation, branching, and synapse formation and ICAM5 is described to selectively target dendrites (25) to endow their protrusions with plastic properties (26).In this work we show that an ICAM5 fusion protein has no detectable effect on neuronal morphology and function and that it is specifically and highly enriched in the somatodendritic compartment of both peripheral nervous system (PNS) and CNS neurons. Next, we use this marker to gain insights into dendrite development, identify dendritic compartments, and map neuronal connections.…”

“…ICAM4 has been reported to be exclusively expressed in erythrocytes and has a suggested role in cell interaction events, including hemostasis and thrombosis (33). ICAM5 is mainly expressed in specific areas of the brain (34) and has been implicated in dendritic outgrowth (35) and rapid cell spreading of microglia (36). Although the reported expression patterns and described functions of ICAM4 and ICAM5 are not indicative of a role in breast and prostate cancer susceptibility, their roles in cell adhesion and cell signaling together with their low level expression in cancer-relevant tissues leave the possibility that their dysregulation or dysfunction may increase cancer risk.…”
