Biosilicification is widespread across the eukaryotes and requires concentration of silicon in intracellular vesicles. Knowledge of the molecular mechanisms underlying this process remains limited, with unrelated silicontransporting proteins found in the eukaryotic clades previously studied. Here, we report the identification of silicon transporter (SIT)-type genes from the siliceous loricate choanoflagellates Stephanoeca diplocostata and Diaphanoeca grandis. Until now, the SIT gene family has been identified only in diatoms and other siliceous stramenopiles, which are distantly related to choanoflagellates among the eukaryotes. This is the first evidence of similarity between SITs from different eukaryotic supergroups. Phylogenetic analysis indicates that choanoflagellate and stramenopile SITs form distinct monophyletic groups. The absence of putative SIT genes in any other eukaryotic groups, including non-siliceous choanoflagellates, leads us to propose that SIT genes underwent a lateral gene transfer event between stramenopiles and loricate choanoflagellates. We suggest that the incorporation of a foreign SIT gene into the stramenopile or choanoflagellate genome resulted in a major metabolic change: the acquisition of biomineralized silica structures. This hypothesis implies that biosilicification has evolved multiple times independently in the eukaryotes, and paves the way for a better understanding of the biochemical basis of silicon transport through identification of conserved sequence motifs.

Solgel methods is a commonly used methods for encapsulation of enzyme and drug, but this method has two disadvantages of using acid or base as a catalyst and being difficult to control pore size of silica material. Even though synthesis under mild condition or silica with controlled pore size in the mesopore region have been reported, it is still difficult to achieve these two characteristics simultaneously. In this work, we chose 10-mer peptides of lysine (K), histidine (H), and block and alternate K and aspartic acid (D) as catalysts for silica mineralization, and silica gels were prepared using the synthetic peptides and a "leave to stand" synthesis method. The resulting silica hydrogels were lyophilized, and their surface areas and morphologies were characterized using the BrunauerEmmettTeller (BET) method and field-emission scanning electron microscopy (FE-SEM), respectively. Silica gels prepared by the "leave to stand" method with K 10 and H 10 exhibited a mesoporous structure with high surface area (576 and 451 m 2 g ¹1, respectively) and pore volume (0.35 and 0.30 cm 3 g ¹1 , respectively). SEM images confirmed the mesoporous structure of these gels. We encapsulated fluorescein sodium salt as a model drug within silica hydrogels using K 10 and H 10 as a catalyst. The silica hydrogel prepared using H 10 exhibited faster release of the drug (approximately 2.5-fold) than gels prepared using K 10 . These results demonstrate that by changing isoelectric point binding between the peptide and pore structure, the synthesized silica hydrogel-peptide composites can be designed to control the release rate of an encapsulated drug.

Some enzymes catalyze the formation of precipitants of metal ions, and some others directly interact with metal-containing precursors. Such enzymes allow the synthesis of inorganic solids under mild conditions. Due to the nature of enzymatic reactions, enzyme-mediated ceramic synthesis techniques can facilitate (1) site-selective ceramic deposition, (2) low temperature ceramic synthesis, (3) intact combination of ceramics with heat-sensitive materials, and sometimes (4) synthesis of metastable phases. The morphology of the resultant ceramics can be tailored in many ways by immobilizing enzymes onto various templates. So far, successful cases have been reported in preparing thin film coatings, hollow micro-or nanospheres, multi-layer stacking, nanotubes, 2D patterns, honeycombs, 3D replicas of biominerals, and so on. Enzyme-mediated ceramic synthesis provoked the emergence of a new type of processing technique which employs proteins or peptides as the mediators. Practical applications of enzyme-or peptide-mediated ceramics are on the horizon in many fields, such as biomaterials, enzyme immobilization supports, and enzyme sensors. This article presents a comprehensive survey of the recent advances of enzyme-mediated ceramic synthesis and related techniques as well as future outlook.
