Interactions between saturated acyl chains confer detergent resistance on lipids and glycosylphosphatidylinositol (GPI)-anchored proteins: GPI-anchored proteins in liposomes and cells show similar behavior.

Abstract: Proteins anchored by GPI are poorly solubilized from cell membranes by cold nonionic detergents because they associate with detergent-resistant membranes rich in cholesterol and sphingolipids. In this study, we demonstrated that cholesterol and sphingolipid-rich liposomes were incompletely solubilized by Triton X-100. GPI-anchored placental alkaline phosphatase incorporated in these liposomes was also not solubilized by cold Triton X-100. As sphingolipids have much higher melting temperatures (Tm) than cellula… Show more

“…Biochemical studies also support this concept and demonstrate that purified cholesterol-rich microdomains isolated from cultured cell (L-cell, MDCK, primary hepatocytes) represent nearly one-third of the plasma membrane, are rich in cholesterol as well as saturated/ monounsaturated fatty acylated phospholipids, and are comprised of physically distinct, liquidordered membrane phases intermediate between fluid liquid-crystalline and rigid gel phases (26,82,102,136,172,182,(215)(216)(217). Studies with purified microdomains from L-cell and MDCK plasma membranes showed that both exogenous (e.g.…”

“…POPC/Sm/GM1/ Chol liposomes have been shown to resemble rafts in several respects, including major lipid composition and low buoyant density, which permits their isolation by flotation through density gradients [14,21]. Finally, we used binary mixtures of POPC with Chol, Sm, or GM1 ganglioside to assess the contribution of individual lipids.…”

“…The difference in detergentsolubility of the DIG proteins at 37 mC between the cerebral cortex membranes and kidney microvillar membranes may well reflect differences in the lipid compositions of these two membranes. Whereas glycosphingolipids are major components of mouse cerebellar membranes [24], they are only minor components of epithelial cell membranes [8], and alkaline phosphatase has been shown to be more insoluble in cerebroside-containing synthetic vesicles than in vesicles of other lipid compositions [45].…”
