The optimum conditions for the activity of the chick liver microsomal oxidative , -dimethylaniline demethylase have been established. Reduced nicotinamide-adenine dinucleotide phosphate was required and was partially replaced by reduced nicotinamide-adenine dinucleotide. The enzyme was competitively inhibited by /3-diethylaminoethyl diphenylpropylacetate. Michaelis constants in the absence and in the presence of /3-diethylaminoethyl diphenylpropylacetate were determined for , -dimethylaniline and , -dimethylaniline A-oxide. The data presented in this work demonstrate that the rate of , -dimethylaniline A-oxide demethylation by chick liver microsomal fraction is sufficiently high for the A-oxide to be considered as an intermediate in the oxidative demethylation of , -dimethylaniline. Ferrous ions activated the microsomal demethylase when incubated with nicotinamide-adenine dinucleotide phosphate and gave a greater oxidative reaction in the absence of nicotinumerous studies have been conducted on the oxidative demethylation of A-methyl compounds. In this paper, we provide an overview of the public health implications of exposure to some pesticides via breast milk and provide health-based guidance. The presence of organochlorine pesticides in breast milk has been documented in many studies around the world. Included in our review are aldrin/dieldrin, chlordane, 1,1,1-trichloro-2,2-bis(p-chlorophenyl) ethane (DDT)/1,1-dichloro-2,2-bis(p-chlorophenyl)ethylene (DDE), endrin, hexachlorobenzene (HCB), and hexachlorocyclohexane (HCH). Toxicological and environmental data on these chemicals are compiled in toxicological profiles published by the Agency for Toxic Substances and Disease Registry (ATSDR). Based on the data, ATSDR derives chemical-specific minimal risk levels (MRLs) that assist in evaluating public health risks associated with exposure. MRLs are health-based guidance values designed to protect the most sensitive populations, including breast-fed infants. We compare MRLs and projected intakes from the breast milk for the listed pesticides, explore the possibilities of toxicological interactions, and provide health-based recommendations.

The properties and actions of the bridged diphenyl acaricides are discussed. These pesticides, which are more or less structurally related to DDT, were the first of the specific acaricides to be developed. They exhibit remarkable properties of specificity, being primarily toxic to phytophagous mites but of very low toxicity to most nontarget species, including insects, fish, birds, and mammals. Although many important facets of their broad mode of action are understood, virtually nothing is known of their primary mode of action or the underlying bases of their specificities. In most ways they are model compounds for integrated control and pest management activities and thus merit greater attention than they have received to elucidate the fundamentals underlying their unusual properties and actions.
