Interaction of silver atomic quantum clusters with living organisms: bactericidal effect of Ag3clusters mediated by disruption of topoisomerase–DNA complexes

Abstract: Ag3 clusters distort DNA conformation altering the binding of doxorubicin and enzymes. As a novel aspect, the Ag3 bactericidal activity is described.

“…This intercalation is strictly dependent on the number of silver atoms in the cluster, being exclusive of Ag 3 ‐AQCS and independent of the type of base pairs (AT of GC) of the double helix . Therefore, it is essential to obtain pure and highly concentrated Ag 3 ‐AQCs without surfactants or other strong binding ligands that could mask their biological activity, which is a substantial challenge. Additionally, the Ag 3 ‐AQC concentration required for this study is roughly two orders of magnitude higher than that of previous studies .…”

“…Cisplatin (line 3) was used as a reference for covalent binding, showing the OC and SC bands migrates more than free pUC18. L4-Ru (lanes 4-6) and L4-Rh (lanes 7-9) cause dosedependant retardation in the migration of supercoiled DNA bands which can be related to intercalation processes [216]. When the amount of L4-Rh is increased, a vanishing of the band intensity takes place, pointing out the precipitation of large amount of Rhodium coordinated to DNA.…”
