“…1) and intercalates between DNA base pairs in solution (25,26). The binding site of anthracyclines covers 3-4 DNA base pairs (27). The high value of ni = 18 + 8 indicates a strong suppression of the conformational flexibility of DNA by the drug.…”

“…From a set of several such titrations, differing in DNA concentration, the equilibrium binding parameters were calculated using the above-mentioned binding model. The corresponding numerical fitting procedure of the orginal titration data was performed for ,B-rhodomycin-I (and recentl y for iremycin (WALTER et al 1983)) with the absorbances a t wavenumber v2 (absorption maximum), whereas for 8-rhodomycin-I1 the measurements at v3 were additionally included. Moreover, to test the internal consistency of the parameter estimations, for @-rhodoniycin-I1 an additional set of data, obtained by titration experiments with independently prepared stock solutions of ligand and of calf-thymus DNA, was used.…”

Table 2 summarizes the equilibrium Table 2 Equilibrium binding parameters of iremycin (WALTER et al 1983, SCIXUTZ et al 1983 ( 1 5 7 ) ( 5 3 6 ( 1 8 7 ) ( 1 5 3 ) DNA binding parameters obtained for the two /I-rhodomycins regarding the dimerization of the ligands. The error limits without parentheses in Table 2 were estimated neglecting the errors of the dimerization parameters from Table 1, whereas those in paraiitheses were obtained with consideration of error propagation.…”
