The VillegasÁGlisson/University of Georgia (VG/GA) strain of Newcastle disease virus (NDV) isolated from the intestine of healthy turkeys has been proposed to replicate in the respiratory and intestinal tract of chickens. In the present study, the virus distribution, the mucosal and systemic immune response, the efficacy against lethal challenge and the full genome sequence of the VG/GA strain were compared with the La Sota strain of NDV. The VG/GA strain was detected at different time points in the respiratory and intestinal tract of chickens with a preferential tropism for the latter. Both the VG/GA and La Sota strains induced NDVspecific immunoglobulin A (IgA) at the upper respiratory tract. IgA levels were higher in the trachea for the La Sota strain, while they were higher in the bile and intestine for the VG/GA strain. Positive correlation between virus distribution of the viruses and IgA production was observed. Despite the presence of the maternal antibodies in broilers, early vaccination with the VG/GA strain afforded 95% to 100% protection against lethal challenge, equivalent to the protection conferred by the La Sota strain. Full genome sequence analysis classified the VG/GA strain within class II, genotype II viruses, which also include most of the respirotropic vaccine strains. Differences with the La Sota strain at the nucleotide and amino acid levels that may explain the differential phenotype of the VG/GA were observed; however, verification of the significance of those changes is required. Taken together, these results validate field observations on the efficacy of VG/ GA vaccination and demonstrated the unique characteristics of the strain.

This paper examines the effects on specific pathogen-free (SPF) chicks when avian metapneumovirus (aMPV) and Newcastle disease virus (NDV) La Sota strain vaccines are co-administered. Day-old SPF chicks were divided into five groups. The first group was inoculated with sterile water (SW) and the rest of the groups were inoculated with live NDV vaccine VG/GA by the oculo-oral route. At 21 days-old, the unvaccinated chicks were again inoculated with SW. The four VG/GA-vaccinated groups were further inoculated with (i) SW, (ii) live aMPV vaccine, (iii) live NDV La Sota, or (iv) combined live NDV La Sota and live aMPV, respectively. Chicks were monitored for post-vaccination reactions and oropharyngeal swabs were collected for vaccines detection. Blood samples were collected to detect aMPV ELISA and NDV haemagglutination-inhibition antibodies. Twenty-one days following the second vaccination, six chicks from each group were challenged with virulent NDV or aMPV respectively. Chicks were monitored for clinical signs and mortality and oropharyngeal swabs collected for aMPV detection. Results showed that, when challenged with a virulent aMPV, both chicks previously vaccinated with VG/GA and subsequently given aMPV vaccine singly or in combination with La Sota were equally protected against clinical signs. Chicks that were vaccinated against NDV either once with VG/GA or followed by La Sota (singly or in combination with aMPV) were fully protected when challenged with velogenic NDV. We concluded that simultaneous administration of live aMPV and NDV La Sota vaccines have no adverse effects on protection conferred by either live vaccine.

Maternally derived antibodies (MDA) don not protect turkeys against rhinotracheitis (TRT) but high MDA influences upper respiratory tract (URT) immunity stimulation after avian Metapneumovirus (aMPV) vaccination. Humoral immunity can not be considered as an indicator of protection against TRT, but specific antibodies inhibit aMPV replication and alleviate the course of TRT. Scarce reports indicate the role of IgA in protection against TRT. The aim of our study was to investigate the impact of MDA on stimulation, antigen specificity acquisition of B lymphocytes, and the production of specific IgA after TRT vaccination of turkeys.The results of our study indicate that MDA on the day of TRT vaccination causes disturbances at different levels of specific humoral immunity expression including antigen specificity acquisition of B IgA + lymphocytes as well as production and secretion of IgA. Vaccine immunity against aMPV associated with sIgA is well expressed in birds not possessing MDA on the day of TRT vaccination, whereas it is inhibited in MDA + birds.These results corroborate our previous findings and indicate that MDA could be responsible for TRT vaccination failure. These findings could explain the observed frequency of TRT field outbreaks despite aMPV vaccination of turkey flocks.
