Interaction between human tRNA synthetases involves repeated sequence elements.

Abstract: Aminoacyl-tRNA synthetases (tRNA synthetases) of higher eukaryotes form a multiprotein complex. Sequence elements that are responsible for the protein assembly were searched by using a yeast two-hybrid system.Human cytoplasmic isoleucyl-tRNA synthetase is a component of the multi-tRNA synthetase complex and it contains a unique C-terminal appendix. This part of the protein was used as bait to identify an interacting protein from a HeLa cDNA library. The selected sequence represented the internal 317 amino acid… Show more

“…A positive interaction was defined as an ability to support cell growth on leucinedepleted media or by a blue result by white color testing on X-galcontaining media (22). DNAs encoding human TB10 WT or T20S mutant (Thr 20 to Ser 20 mutation) for in vitro binding assay was ligated into pGEX4T-1 (Amersham Biosciences) using EcoRI and SalI to generate glutathione S-transferase (GST) fusion protein.…”

“…Ovarian Cancer CellsWe tested the contribution made by individual amino acids in the 17 LKKTET 22 motif to the biological activity of TB10 by Ala and Ser scanning mutagenesis. Each of the mutant sequences was confirmed by automatic sequencing, and transfected to SKOV-3 and PA-1 (data not shown) ovarian cancer cells.…”

“…Given the variety of functions that have been associated with aminoacyl-tRNA synthetases (46), sumoylation of these enzymes could help mediate the stress response in any number of ways. For example, both EPRS and MRS are components of the MSC, and the rep domain, which is the region of EPRS targeted for sumoylation, is required for the formation of this complex (67). The functional significance of the MSC is unclear, although it may play a role in the trafficking of tRNA from the nucleus to the ribosome (47).…”

“…The underlying basis of association has been demonstrated for several of the polypeptides to be the hydrophobic nature of extensions unique to the eukaryotic enzymes (reviewed in Mirande, 1991;Yang, 1996). In addition, specific sequence motifs have been identified in synthetase polypeptides and p l 8 that play a role in proteinprotein interaction (Quevillon & Mirande, 1996;Rho et al, 1996). The overall shape and size of the particle are known from electron microscopy studies (Norcum, 1989).…”
