Interaction between HLA-B60 and HLA-B27 as a Better Predictor of Ankylosing Spondylitis in a Taiwanese Population

Abstract: ObjectiveAnkylosing spondylitis (AS) is a form of chronic inflammatory spondyloarthritis (SpA) that causes pain and stiffness in spines or joints. Human leukocyte antigen B27 (HLA-B27) and B60 (HLA-B60) have been reported as major genetic risk factors of AS. In addition, rs13202464, located on major histocompatibility complex (MHC) region, showed high sensitivity (98.7%) and specificity (98.0%) for HLA-B27.DesignThe aim of our study is to test whether the interaction between HLA-B60 and HLA-B27 (rs13202464) ca… Show more

“…This epistatic (gene/gene) interaction suggests that ERAP1 contributes to disease susceptibility by exacerbating the pathogenic role of HLA-B*27. Another epistatic interaction results in a greater risk for developing AS in patients heterozygous for two HLA class I risk alleles (HLA-B*27 and HLA-B*40:01, the latter encoding the Bw60 serotype) than would be expected from the additive effect of the two alleles on disease risk 100,101 .…”

“…Studies of the contribution of major histocompatibility complex (MHC)-encoded variants to the heritability of ankylosing spondylitis (AS) suggest that it is responsible for 20.44% of the genetic risk for the disease, with over 114 non-MHC variants identified to date contributing another 7.38% 1. Other MHC region genes have also been implicated in addition to HLA-B*27 , including HLA-B*14 ,2 3 HLA-B*60 ,4–8 HLA-DRB1, 9–11 HLA-DPB1, 11 12 MICA ,13 14 TNF-alpha1 ,15 TAP1 16 and LMP2, 11 17 although the level of association of these findings has not been definitive and with the exception of HLA-B*60 replication has not been universal.…”
