Inter-regulation of IGFBP1 and FOXO3a unveils novel mechanism in ursolic acid-inhibited growth of hepatocellular carcinoma cells

Abstract: BackgroundUrsolic acid (UA), a natural pentacyclic triterpenoid, exerts anti-tumor effects in various cancer types including hepatocellular carcinoma (HCC). However, the molecular mechanisms underlying this remain largely unknown.MethodsCell viability and cell cycle were examined by MTT and Flow cytometry assays. Western blot analysis was performed to measure the phosphorylation and protein expression of p38 MAPK, insulin-like growth factor (IGF) binding protein 1 (IGFBP1) and forkhead box O3A (FOXO3a). Quanti… Show more

“…While PPARc is upstream of many of the effectors that are influenced by BAR, the PPARc agonist rosiglitazone did not recapitulate the effects of BAR on gene expression or prevent loss of metabolic activity. Others have shown that FOXO3a is a target for triterpenoids in cancer [15,37].…”

“…Yang et al30 reported that “UA significantly arrested cycle in G0/G1 phase of hepatocellular carcinoma cells”. However, the underlying mechanism of synergized antiproliferative effects of UA–cisplatin on cancer cells is not completely understood.…”

“…High expression of IGFBP mRNA was strongly correlated with better survival in breast cancer mouse model [30]. We previously observed that ursolic acid, a natural pentacyclic triterpenoid, and emodin, one anthraquinones constituents derivative isolated from the roots of rheum palmatuma, inhibited growth of hepatocellular carcinoma and lung cancer cells through induction of IGFBP1 gene expression, suggesting tumor suppressing role of this molecule [31, 32]. However, conflicting results were observed in other cancer types, such as prostate [33, 34] and endometrial cancer [35] and others [36].…”
