Ultrasound-assisted extraction (UAE) is widely used; however, the efficiency of extraction depends on the raw materials. Therefore, optimization of UAE must be investigated for each type of plant material. By-products from soursop fruit have not been studied as a source of bioactive compounds. In this work, the optimization of UAE conditions (extraction time (5, 10, and 15 min), pulse cycle (0.4, 0.7, and 1 s), and sonication amplitude (40%, 70%, and 100%)) for the extraction of phenolic compounds (soluble, hydrolyzable, condensed tannins, and total polyphenols) from soursop by-products (seed, peel, and columella) and pulp was evaluated using response surface methodology. The optimal conditions for UAE to obtain the highest total polyphenol content from by-products and pulp was dependent on the raw material. Peel resulted in the highest content of total polyphenols (187.32 mg/g dry matter [DM]) followed by columella (164.14 mg/g DM), seed (36.15 mg/g DM), and pulp (33.24 mg/g DM). The yield of polyphenolic content from peel and columella obtained with UAE was higher (32–37%) than conventional extraction for 2 h under stirring (14–16%). The contents of gallic acid (0.36–15.86 µg/g DM), coumaric acid (0.07–1.37 µg/g DM), and chlorogenic acid (9.18–32.67 µg/g DM) in the different parts of the fruit were higher in the extracts obtained by UAE compared with a conventional extraction method (0.08–0.61, 0.05–0.08, 3.15–13.08 µg/g DM, respectively), although it was dependent on the raw materials. Soursop by-products can be functionally important if they are used to extract bioactive compounds by UAE; a technology with high potential for commercial extraction on a large scale.

Antioxidant compounds were produced from submerged fermentation using Botryosphaeria dothidea. Sonication times (1-15 min) and agitations (0-150 rpm) applied to different fermentation phases (2-7 days) were evaluated for their effect on the broth antioxidant activity. In approximately 70% of assays performed with ultrasound, the antioxidant activity of the broth containing exocellular metabolites was higher than the control (without sonication), reaching a maximum value of 96%. The solid fraction (biomass after supernatant removal) corresponding to the best assay of antioxidant activity was submitted to extractions with water, ethanol or ethyl acetate to recover intracellular metabolites. The yields of extracts were 3.7 ± 2.6 wt %, 20.6 ± 3.9 wt % and 32.0 ± 4.2 wt % for ethyl acetate, ethanol and water, respectively. The use of ethanol could provide a larger number of compounds such as pentadecanoic acid, ergosta-5,8,22-trien-3-ol, and 1,4-diaza-2,5-dioxobicyclo [4.3.0] nonane. Both the supernatant and solid fractions revealed some bioactive compounds with promising antioxidant activity with the potential to replace some synthetic antioxidants.

Extraction of tannin Rhizophora Mucronata barks by using two methods, which were soxhlet extraction and boiling were demonstrated in this study in order to evaluate the effective technique for extraction. Several parameters such as the type of solvent used, time of extraction, and sample size were specified. The extractant was then evaporated using vacuum rotary evaporator. The percentage of extractives and the characterization of condensed (Stiasny Index) and hydrolyzable tannins were all quantified. The milled barks with different sizes were introduced to the extraction using water, methanol, acetone, and toluene as a solvent for three, six and nine hours for Soxhlet extraction while for boiling only water were used as a solvent and at four, six and eight hours. Results demonstrated that extraction from the bark of Rhizophora Mucronata with water for six and nine hours extracted approximately 29 wt% and 13wt % for both soxhlet extraction and boiling respectively when coupled with vacuum rotary evaporator. The Soxhlet extraction method was superior as compared to boiling. The optimum parameters for the Soxhlet extraction were water as the solvent at six hours extraction with 0.5 mm of milled bark particles size that extracted 29 wt% of extractives. The condensed tannin and hydrolyzable tannin of the extractives using soxhlet extraction were approximately 76 wt% and 0.00133 wt% respectively.
