Intelligent Photosensitive Mesenchymal Stem Cells and Cell-Derived Microvesicles for Photothermal Therapy of Prostate Cancer

Abstract: Targeted delivery of nanomedicines into the tumor site and improving the intratumoral distribution remain challenging in cancer treatment. Here, we report an effective transportation system utilizing both of mesenchymal stem cells (MSCs) and their secreted microvesicles containing assembled gold nanostars (GNS) for targeted photothermal therapy of prostate cancer. The stem cells act as a cell carrier to actively load and assemble GNS into the lysosomes. Accumulation of GNS in the lysosomes facilitates the clos… Show more

“…A similar approach to the one described by the Aboody group was performed on modified MSCs by loading them with gold nanoparticles (nanostarts) to improve phototherapy. Although studied in a heterotopic model of prostate cancer, the results support the use of MSC to maximize clinically relevant gold nanoparticles’ optical–electronic properties by increasing nanoparticle intratumor distribution ( Huang L. et al, 2019 ).…”

“…In particular, several authors have shown that MSCs easily internalize different types of nanoparticles [27] and can reach the tumor, showing limited or no toxicity effect of NPs to MSCs [58,59]. Additionally, other authors have investigated whether MSCs can transport nanoparticles for therapeutic purposes [22,60,61].…”

“…Another valuable tool for targeted PTT of prostate cancer is so-called mesenchymal stem cell-derived gold star nanoparticles produced directly in the cells and secreted in a form of microvesicles based upon a light irradiation stimulus. The main advantage of such nanoparticles is the fact that they exhibit up to 90% efficacy of the photothermal transformation [62,63].…”
