Integrin Fibronectin Receptors in Matrix Metalloproteinase-1–Dependent Invasion by Breast Cancer and Mammary Epithelial Cells

Abstract: Integrins contribute to progression in many cancers, including breast cancer. For example, the interaction of ␣ 5 ␤ 1 with plasma fibronectin causes the constitutive invasiveness of human prostate cancer cells. Inhibition of this process reduces tumorigenesis and prevents metastasis and recurrence. In this study, naturally serum-free basement membranes were used as invasion substrates. Immunoassays were used to compare the roles of

“…Integrins are the most important matrix receptors and their downregulation can promote dissemination of primary tumor but also suppress the attachment of already migrating tumor cells at distant sites. In accordance, downregulation is crucial for the establishment of invasive phenotype in breast cancer (50). Downregulation of ITGA4 in PR9692-E9 cells leading to inhibition of metastatic dissemination can be an example of the second mechanism.…”

“…Using a model system for invasion that utilizes a naturally occurring membrane found in sea urchin embryos (Livant, Linn et al 1995) in the configuration a cell would see upon extravasation (FIGURE 3), we determined that 5 integrin binding to the PHSRN sequence of fibronectin is necessary for invasion and that 4 integrin binding to the "LDV" sequence of fibronection abrogates invasion (Livant, Allen et al 2000;Livant, Brabec et al 2000;Woods Ignatoski, Maehama et al 2000;Jia, Markwart et al 2002) (FIGURE 4). Using this system, we showed that HER-2 requires PI3'K to drive invasion.…”
