Integrative Transcriptomic, Proteomic and Functional Analysis Reveals ATP1B3 as a Diagnostic and Potential Therapeutic Target in Hepatocellular Carcinoma

Abstract: The Na+/K+-ATPase (NKA), has been proposed as a signal transducer involving various pathobiological processes, including tumorigenesis. However, the clinical relevance of NKA in hepatocellular carcinoma (HCC) has not been well studied. This study revealed the upregulation of mRNA of ATP1A1, ATP1B1, and ATP1B3 in HCC using TCGA, ICGC, and GEO database. Subsequently, ATP1B3 was demonstrated as an independent prognostic factor of overall survival (OS) of HCC. To investigate the potential mechanisms of ATP1B3 in H… Show more

“…β3 is involved in cell proliferation through the PI3K/AKT (phosphatidylinositol 3-kinase/protein kinase B) signaling pathway, and cells in which β3 is knocked down tend to undergo apoptosis ( Li et al , 2017 ). Similar reports on hepatocellular carcinoma suggest that β3 may be a biomarker for prognostic prediction ( Lu et al , 2021 ). These reports suggest that β3 may play a role during the adaptation of cells to environments that differ from the in vivo environment or that require higher energy efficiency.…”

“…Na+/K+-ATPase (NKA) has been proposed as a signal transducer involved in various pathobiological processes, including hepatocellular carcinoma (HCC) ( 33 ). We identified an enrichment of the ATP1B1 an astrocyte-specific isoform of the Na+/K+-ATPase transmembrane ionic pump, by RNA sequencing analysis of endothelial cells from a mouse liver fibrosis model ( Figure 5A ).…”

“…To date, 4 NKA a-subunits (a1, a2, a3, and a4) and 4 b-subunits (b1, b2, b3, and b4) have been identified. Abnormal NKA can lead to a variety of diseases, including hypokalemic periodic paralysis and CNS symptoms ( 47 ), cardiovascular disorders ( 48 ), atherosclerosis ( 49 ), Alzheimer’s disease ( 50 ), and hepatocellular carcinoma (HCC) ( 33 ). ATP1B1 is downregulated in human epithelial cancer cells ( 51 53 ).…”

“…In addition, several other PSGs ( ATP1B3 , TFDP2 , GK5 , XRN1 , PEX5L , USP13 , NDUFB5 , MRPL47 , ACTL6A , KIF26B , CDKL5 , and HDAC4 ) were identified by other four selective signal analyses and DCMS ( Supplementary Figures S4, S5 ). ATP1B3 is related to immune cell infiltration and immune-related cytokines expression in hepatocellular carcinoma ( Lu et al, 2021 ). TFDP2 affects angiogenesis, nephrogenesis, and kidney metabolic function ( Köttgen et al, 2010 ).…”
