Integration of target discovery, drug discovery and drug delivery: A review on computational strategies

Abstract: Most of the computational tools involved in drug discovery developed during the 1980s were largely based on computational chemistry, quantitative structureactivity relationship (QSAR) and cheminformatics. Subsequently, the advent of genomics in the 2000s gave rise to a huge number of databases and computational tools developed to analyze large quantities of data, through bioinformatics, to obtain valuable information about the genomic regulation of different organisms. Target identification and validation is a… Show more

“…NCs consisting of the triblock terpolymers PVFc-b-poly(methyl methacrylate)-b-poly(N,N-dimethylaminoethylmethacrylate) show triggered release of the payload under three different stimuli, namely, pH change, oxidizing agent, and temperature. 247 Redox/pH dual-responsive supramolecular vesicles based on the host-guest complexation of Fcium carboxylic acid capped pillar [5]arene (FACP5) and a galactose derivative (G) were used for targeted drug delivery (Fig. 38).…”

“…Drug delivery is the most promising approach to solving the problem of the safe transportation of a drug through the body to achieve its target. 4,5 Targeted drug delivery will solve several problems: protect drugs from degradation and unwanted interactions with biological molecules, increase the selective absorption of drugs by tumor cells, monitor pharmacokinetics, and increase the bioavailability of drugs inside tumor cells. Targeted delivery combined with controlled drug release plays a key role in the future of personalized medicine.…”

“…In the present study, we evaluated the antiviral effects of curcumin derivatives as potent inhibitors of influenza H1N1 neuraminidase based on 3D quantitative structure-activity relationship (QSAR) (Duarte et al 2019), molecular docking (Nilakantan et al 1993) and biological activity evaluation both in vitro and in vivo.…”

“…They were mainly based on structureactivity relationship. The drug discovery system becomes the most important approach towards the targeted cells [11]. Traditional antisense and RNA interference (RNAi) technologies are the robust tools used in multidimensional phases to discover and validate the potential drug targets.…”
