Integration of a barley (Hordeum vulgare) molecular linkage map with the position of genetic loci hosting 29 developmental mutants

Abstract: The first step in positional gene cloning is the integration into available molecular maps of genetic loci for which mutant alleles exist. We report the placement of 29 barley developmental mutants on a restriction fragment length polymorphism-amplified fragment length polymorphism (RFLP-AFLP) map. The mapping procedure used homozygous mutant F 2 plants in an iterative process: once a mutant linked AFLP was found, primer combinations were successively selected to generate AFLP fragments more tightly linked to … Show more

“…Previous mapping positioned Cul4 on the distal end of chromosome 3HL (Pozzi et al, 2003;Druka et al, 2011). High-resolution mapping using 9,898 gametes from the cross cv Bowman-cul4.5 3 cv Morex located Cul4 to a 0.55-centimorgan (cM) interval (Fig.…”

“…In some indica rice varieties, the ERF VII gene Submergence1A and related SNORKEL1 (SK1) and SK2 genes have been shown to play important (and contrasting) functions in ethylene-mediated internode growth in response to submergence (Hattori et al, 2009;Bailey-Serres and Voesenek, 2010;Hinz et al, 2010;Jung et al, 2010). a As defined by Castiglioni et al (1998) and Pozzi et al (2003). Transcriptional regulation by ethylene has been reported for other ERF VII genes, including OsEBP89/ OsERF60 and OsEBP2/OsERF72 in rice (Yang et al, 2002;Mao et al, 2006;Lin et al, 2007), RAP2.2 in Arabidopsis (Hinz et al, 2010), and HvRAF (for H. vulgare root abundant factor), the only characterized member of this group in barley (Jung et al, 2007).…”

“…Moreover, it is known that the very large majority of segregating AFLP fragments map at biallelic loci. Our laboratory has located 438 such loci on an einkorn linkage map (Taenzler et al 2002) and 819 on an integrated map of barley (Pozzi et al 2003). In barley, it has been clarified that comigrating AFLP bands produced by amplifying DNA of different genotypes derive from identical DNA sequences.…”
