Integrating optical tweezers with up-converting luminescence: a non-amplification analytical platform for quantitative detection of microRNA-21 sequences

Abstract: We report a single-microsphere based imaging assay method by integrating up-converting luminescence with optical tweezers for detecting microRNA-21 sequences. This method achieves a competitive detection limit of 12 fM with good selectivity and no dedicated signal amplification designs.

“…After the targets were enriched and the mentioned quantitative analysis was undertaken, it was determined that each case exhibits different amounts of miRNA-21 sequences at the single cell level (Figure B), i.e., 1878, 603, and 162 copies/cell for MCF-7, HepG2, and Jurkat T, respectively. The conspicuous difference is thought to be due to the vital relevance between miRNA-21 and breast cancer . Meanwhile, the quantified data are evidenced to agree with the real-time PCR (RT-PCR) analysis (Figure S16 and Figure C), demonstrating the reliability of the proposed analytical technique.…”

“…Based on the above evidence, we were inspired to integrate OT with specially designed luminescence confined UCNPs to pave a new way to a significantly enhancing bead-supported detection method. The assay principle is illustrated in Scheme , where a kind of next-generation biomarker (miRNA-21 sequences) , for breast cancer is selected as the target model. Concretely, the innovative sandwich structured UCNPs (SUCNPs) with blue emission at around 480 nm are first fabricated to wholly restrict the lanthanide activators (Tm 3+ ) into the inner shell with an extremely narrow space range (≈ 3 nm).…”

“…Logically, two key points should be considered before putting the proposed suspension array into effect. Unlike our previous work by using a single NIR beam to trap 3 μm beads, , in this study we attempt to split the same laser beam to generate an array of 3 × 3 traps. Thus, it is critical to calibrate whether this case is robust enough to implement multiple trapping.…”

“…Liang et al combined optical tweezers and “light knife” to perform surgery on cells and organelles, including suspension, movement, sorting, and fusion, etc., and made quantitative measurement of the internal mechanical properties of cells and the interaction between cells [ 6 ]. Li and Tang et al successfully reported the sorting [ 7 ] and sequencing [ 8 ] of a single chromosome. The rigidity caused by molecular shrinkage has also been accurately characterized by optical tweezers [ 9 ].…”
