Integrated single-cell RNA-seq analysis identifies immune heterogeneity associated with KRAS/TP53 mutation status and tumor-sideness in colorectal cancers

Abstract: BackgroundThe main objective of this study was to analyze the effects of KRAS/TP53 mutation status and tumor sideness on the immune microenvironment of colorectal cancer using integrated scRNA-seq data.MethodsA total of 78 scRNA-seq datasets, comprising 42 treatment-naive colorectal tumors, 13 tumor adjacent tissues and 23 normal mucosa tissues were included. Standardized Seurat procedures were applied to identify cellular components with canonical cell marks. The batch-effect was assessed and corrected using … Show more

“…Initially, T cells need high levels of OXPHOS to maintain cell growth homeostasis and differentiation integrity, and following activation, Teffs exhibit reduced mitochondrial respiration rates, whereas glycolysis and glutamine metabolism increase to ensure an efficient energy supply 68–70 . This observation has been validated using scRNA‐seq data together with lifetime imaging of endogenous fluorescence factors 9,71 .…”

“…According to the DEGs, LYVE1 attracted our attention. LYVE1 is widely expressed on the lymphatic endothelium, in subsets of vascular endothelial cells, and on some subgroups of macrophages, such as tumor-in ltrated PvTAMs [17]. The molecular functions of LYVE1 are diverse.…”

“…The patterns of gene expression and heterogeneity identified through bulk RNA-seq and scRNA-seq have often not been validated experimentally [ 70 ]. We first examined and confirmed to be highly expressed in CRC tissues by immunohistochemistry, RT-qPCR, and western blotting.…”
