Integrated microfluidic device for the separation, decomposition and detection of low molecular weight S-nitrosothiols

Abstract: S-nitrosothiols (RSNOs) are electrokinetically separated and the nitrite generated by decomposition with mercury(ii) is detected by amperometry, in an integrated microdevice.

“…For facilitating the photolysis of nitrosothiol groups and realizing the amperometric detection of nitrite, the authors reported a simple degradation through UV-LED and an analysis with a procedure time shorter than 2 min and LOD value of 73 nmol L –1 . As above-discussed, the proposed modified PES provides suitable detectability for CySNO and the estimated LOD is better or similar when compared to low-cost analytical tools. Also, the herein reported linear concentration range (10–125 μmol L –1 ) can be useful to clinical application that involved Parkinson’s disease …”

“…The achieved LOD was compared to other values found in recent published reports. Duarte-Junior et al 65 proposed a microfluidic device with integrated electrochemical detector for separating and detecting RSNOs, after a decomposition to nitrite based on mercury with LODs of 20 μmol L −1 . Ismail et al.…”

“…Therefore, a buffer solution composed of lactic acid and histidine was prepared at different ratios to investigate the pH effect on nitrite determination. Th analyses were carried out in an acidic medium to favor nitrite oxidation [ 65 , 66 , 67 , 68 ]. For this purpose, the lactic acid concentration was fixed at 30 mmol L −1 and the histidine concentration ranged from 5 to 25 mmol L −1 (5 mmol L −1 increments), providing solutions with pH at 3.2, 3.6, 3.8, 4.1, and 4.5, respectively (a–e) ( Figure 2 A).…”
