Integrated analysis of promoter mutation, methylation and expression of AKT1 gene in Chinese breast cancer patients

Abstract: BackgroundAs downstream mediators of PI3K /PTEN /AKT /mTORC1 pathway, the AKT isoforms play critical roles in tumorgenesis. Although the pleiotropic effects of AKT1 in breast cancer have been reported, the genetic and epigenetic characteristics of AKT1 promoter region in breast cancer remains to be identified. In this study we aimed to investigate the promoter mutation spectrum, methylation and gene expression pattern of AKT1 and their relationship with breast cancer.MethodsBy using PCR target sequence enrichm… Show more

“…Nevertheless, many authors have employed the strategy where genes associated with both altered gene expression and methylation are sought, to identify biologically important genes. Heng et al [ 43 , 44 ] have tried to find genes associated with both altered gene expression and methylation to discover genes crucial for breast cancers. Li et al [ 45 ] have attempted to find genes associated with both altered gene expression and methylation to identify key genes in severe oligozoospermia.…”

“…Moreover, M-protein is responsible for apoptosis in the host cell through inhibition of transcription through host RNA polymerase dumping as well as interruption of nucleocytoplasmic transport of RNAs. 20 , 21 , 22 Researchers have identified wild M-protein as the key harmful molecule for infected cells. 23 …”

“…Gene promoter regions are essential sites in DNA recognized by proteins for the downstream processes of replication and transcription [ 12 , 13 ]. Mutations in promoter regions, as a generalization, can often lead to down or up-regulation of genes that may lead to malignancies [ 14 , 15 , 16 , 17 , 18 ]. For example, in gastric adenocarcinomas, specific mutations in the AKT1 promoter region have been identified from clinical samples and confirmed in vitro to upregulate the Akt/PKB (protein kinase B) pathway, a central pathway for cancer cell proliferation [ 17 , 18 ].…”
