Integrated Analysis of LncRNA-Mediated ceRNA Network in Calcific Aortic Valve Disease

Abstract: Background: The high morbidity and mortality of calcific aortic valve disease (CAVD) represents an unmet clinical need to investigate the molecular mechanisms involved. Evidence suggests that long non-coding RNAs (lncRNAs) can act as competitive endogenous RNAs (ceRNAs) by binding to microRNAs and regulating target genes in cardiovascular diseases. Nevertheless, the role of lncRNAs related ceRNA regulation in CAVD remains unclear. Methods: RNAseq data of human diseased aortic valves were downloaded from GEO da… Show more

“…The ECM is extensively involved in the regulation of miRNAs and transcription factors. [ 21 ] To explore miRNA- and transcription factor-mediated regulation of ECM genes, a gene-miRNA and a TF-gene interaction network were predicted and constructed. Each gene corresponded to an individual miRNA and transcription factors, and partial miRNAs and transcription factors were linked to different genes.…”

“…Studies clarified that miR-26a can promote angiogenesis under various physiological and pathological conditions [17,18]. Previous reports have displayed that lncRNAs had considerable impact on miRNA-mediated gene downregulation or upregulation by playing the role of competing endogenous RNAs or miRNA sponges [19][20][21]. Using bioinformatics tools, we detected the binding site for miR-26a in TUG1.…”

“…The mechanisms underlying the pathogenesis of CAVD remain elusive, and this disease lacks effective drugs [4]. There is an unmet need to investigate the molecular mechanisms of CAVD in face of high morbidity and mortality [5]. In this context, identification of possible future molecular targets may open up new opportunities for the prevention and management of this debilitating disease.…”
