Integrated Analysis of Global mRNA and Protein Expression Data in HEK293 Cells Overexpressing PRL-1

Abstract: BackgroundThe protein tyrosine phosphatase PRL-1 represents a putative oncogene with wide-ranging cellular effects. Overexpression of PRL-1 can promote cell proliferation, survival, migration, invasion, and metastasis, but the underlying mechanisms by which it influences these processes remain poorly understood.MethodologyTo increase our comprehension of PRL-1 mediated signaling events, we employed transcriptional profiling (DNA microarray) and proteomics (mass spectrometry) to perform a thorough characterizat… Show more

“…To assess LCK autophosphorylation activity, we expressed full-length LCK tagged with Myc in HEK293 cells, which lack endogenous LCK (62), and monitored the production of the protein by Western blot with an antibody against the Myc epitope and the amount of phosphorylated LCK with an antibody recognizing LCK phosphorylated at Tyr 394 , which is in the activation loop (Fig. 4B).…”

“…Finally, over-expression of eIF6 in primary melanoma cell lines (WM793) induced cdc42 up-regulation and increased motility and invasiveness, thus demonstrating that the tumor-promoting ability of this initiation factor is not restricted to the A2780 cell line. Notably, both eIF6 and cdc42 have been reported to be up-regulated in cells over-expressing PRL-1, a putative oncogene involved in the control of a number of diverse biological processes, including migration and invasion [ 31 ]. Our results could suggest a possible mechanism of regulation in which eIF6 act as a mediator of the cdc42 expression at the translational level, although additional experiments are needed to elucidate this issue.…”

“…Therefore, a comprehensive understanding on biological systems will require extending single omics further to multi‐omics analysis for gene therapy research. Multi‐omics studies have been conducted in other biologics research (Ang et al, 2019; Dumaual et al, 2013; Shin et al, 2018; Stolfa et al, 2018). Similarly, multi‐omics can be applied to understand the challenges and bottlenecks for rAAV production, such as issues in low productivity, scale‐up, and establishing stable producers.…”
