Insulin and the insulin receptor collaborate to promote human gastric cancer

Abstract: Background Gastric adenocarcinoma is common and consequent mortality high. Presentation and mortality are increased in obese individuals, many of whom have elevated circulating insulin concentrations. High plasma insulin concentrations may promote, and increase mortality from, gastric adenocarcinoma. Tumour promotion activities of insulin and its receptor are untested in gastric cancer cells. Methods Tumour gene amplification and expression were computed f… Show more

“…The relevance of IR to cancer has been demonstrated in clinical studies. Insulin, as a peptide hormone that stimulates tissue accretion, has a cancer-promoting effect [ 78 , 79 ]. Karlstad et al demonstrated that insulin use increases the risk of lung cancer [ 80 ].…”

“…In a previous case-control study, early GC was associated with diastolic blood pressure, total cholesterol, fasting glucose, and the homeostasis model assessment of the insulin resistance (HOMA-IR), which directly showed an association between insulin resistance and GC 34 . In addition, a recent experimental study showed that insulin and insulin-like growth factor (IGF) signal transduction pathway was dominant in gastric adenocarcinoma and responsible for the proliferation and survival of cancer cells 35 . Similarly, markers of activated IGF pathways have been shown to be prognostic factors for patient with GC 36 , which may be alternative evidence that hyperinsulinemia and insulin resistance are related with poor prognosis of patients with GC.…”

“…In addition to the observational evidence, many experimental studies provided solid confirmation of the positive relationship between insulin and cancer. Saisana et al reported that insulin has direct tumor-promoting effects on the gastric adenocarcinoma cells [ 56 ]. In the case of breast cancer, evidence revealed that insulin stimulates the proliferation of some human breast cancer cell lines in vitro by activating both the PI3K/Akt and the MAPK signaling pathways [ 57 ].…”
