Spermathecae of the mangrove crab Ucides cordatus: a histological and histochemical viewUcides cordatus is the most commercially important mangrove crab in Brazil. In spite of its economic importance, there are few studies of its reproduction, in particular the female reproductive system. The present study describes the histology and histochemistry of the spermathecae of U. cordatus. Adult females were caught monthly from July 2004 through June 2005, at Iguape, State of São Paulo. The crabs were anaesthetized, and their spermathecae removed and fixed in Davidson's fluid, following the histological routine for paraffin. The slides were stained with HE, xylidine Ponceau, PAS, alcian blue (pH 1.0 and 2.5), Sudan black B and picrosirius-haematoxylin. Histologically, the spermathecae possesses a capsule of conjunctive tissue, rich in collagen fibres, which surrounds the secretory columnar epithelium. In the lumen, individual sperm packets are not observed; the spermatophores are intermixed with the seminal fluid and secretions of the spermathecae itself. A large proportion of the free spermatozoids and spermatophores are arranged in homogeneous masses in the proximal part of the spermathecae. The secretion produced by the columnar epithelium appears to promote the movement of the gametes to the fertilization chamber, in a ventral position, allowing fertilization of the oocytes. Histochemically, the secretion produced by the columnar epithelium was strongly positive for neutral polysaccharides, positive for acid polysaccharides, and weakly positive for proteins and lipids. This secretion forms a glycoprotein matrix which is associated with maintenance of the spermatophores, which can remain stored for long periods.

(Lepidoptera: Bombycidae) RESUMO PALAVRAS-CHAVE: Insecta, bicho-da-seda, Baculoviridae, lamela neural, perineuro ABSTRACT -BmMNPV, a Nucleopolyhedrovirus isolated from infected Bombyx mori (L.) larvae in Paraná State -Brazil, was used to inoculate healthy 5 th -instar B. mori larvae and examine the infection on central nervous system (CNS) cells. Samples of nervous tissue were removed from the infected insects, at different sampling times, and processed for cytopathology studies by light and transmission electron microscopy using routine techniques. The experiment included both inoculated and noninoculated larvae (control). BmMNPV infection was detected on the 5th day after inoculation in CNS cells. Initially, infection was characterized by nuclear hypertrophy and the presence of virogenic stroma, in which the progeny virions were produced. Virions are enveloped and occluded into protein crystal, the polyhedra. Lyses of infected CNS cells were undetected; however, free mature polyhedra were seen in spaces inside the CNS. These polyhedra possibly came from trachea that penetrate the CNS and its cells, which are susceptible to BmMNPV and lyses after infection. We conclude that the tracheal system is responsible for disseminating BmMNPV infection in B. mori CNS and that the tracheal branches allow non-occluded virions to pass through the blood-brain barrier.

ABSTRACT. The gonadal structure and gametogenesis of Loricaria lentiginosa Isbrücker, 1979 were studied through anatomical and histological techniques. Forty two males and ten females in maturation/mature stage were captured in the reservoir of Porto Colombia, Paraná river basin, Minas Gerais, using gill nets, from November 2001 to October 2002 (tri-monthly captures). Examination of the testes revealed that they were paired, elongated and not fringed organs. Histologically, the testes presented three distinct regions: cranial espermatogenic; transistion espermatogenic and secretory; and caudal exclusively secretory. Standard histochemical techniques detected neutral glycoproteins from the secretion of the tubules of the caudal region. Espermatogenesis occurred in cysts throughout the whole extension of the wall of the seminiferous tubules, which anastomosis themselves and liberated the spermatozoa into the lumen of the espermatic ducts. The ovaries were paired, saculiformes and, histologically, they presented ovigerous lamellae that contained the cells of ovogenic ancestry. The oocytes were classified into four stages, based on their cytological characteristics and the cell layers that surrounded them. Post-ovulatory follicles and vitelogenic oocytes in the follicular atresia process also were observed.
