The musk gland in an adult male forest musk deer is an organ that synthesizes, stores, and secretes musk, a cream-colored liquid upon initial secretion that gradually transforms into a blackish-brown solid substance upon full maturation. In this study, four healthy adult male forest musk deer were selected and a total of 12 musk samples were collected for analysis. The samples were in three different states depending on the different seasonal collection dates, which were in June, August, and October. High-throughput 16S-rRNA gene sequencing technology was used to detect microbiota changes in the gland. The results indicate that microbial richness gradually declined during the musk maturation process. The microbiota composition between the initial liquid and final solid musk samples was varied significantly (P < 0.05). The dominant bacterial phyla were similar at all three stages included Firmicutes, Proteobacteria, Actinobacteria, and Bacteroidetes. However, the abundances were differences in terms of the dominant bacterial genera. PICRUSt analysis showed the highest represented category was “Amino acid transport and metabolism” (24.8%), followed by “Transcription” (22.04%), and “Carbohydrate transport and metabolism” (20.74%). Our findings indicate that the microbiota in the musk gland plays an important role in the maturation process of musk.

Background: While lung cancer poses a serious threat to human health, non–small cell lung cancer (NSCLC) is the most common type of lung cancer. Danggui Buxue Decoction (DBD) is a classic traditional Chinese medicine (TCM) commonly prescribed for anti-tumor use in China. However, the potential mechanism of DBD against NSCLC has not yet been expounded. Hence, this study clarified its potential mechanism through bioinformatics technology.Methods: First, the active ingredients of DBD and the relevant targets were searched and screened by using the TCMSP database. Then the targets of NSCLC were obtained through searching GeneCards and OMIM database. Venny diagram was drawn to find the core targets, and an ingredient-disease-target network diagram was made by Cytoscape software, followed by GO functional enrichment analysis and KEGG pathway enrichment analysis via R software, and molecular docking via the Autodock Vina software , respectively. Finally, a survival analysis was performed to evaluate the prognosis of NSCLC patients through key genes.Results: A total of 22 active ingredients of DBD were obtained. The targets found were 182 relevant to DBD and 5,773 to NSCLC, respectively, among which 140 common targets were obtained from Venny diagram. According to the enrichment degree value, five main active ingredients and ten core targets were identified. The enrichment analysis further revealed that DBD could achieve the purpose of treating NSCLC through the AGE-RAGE signaling pathway in diabetic complications. The findings from molecular docking demonstrated that quercetin and hederagenin have the best working mechanisms with PDE3A and PTGS1, and those from the survival analysis indicated that high PDE3A gene expression has a relatively poor prognosis for NSCLC patients (p < 0.05), PDE3A may be mainly distributed in the LU65 cell line originated from Asian population. Conclusions: The present study demonstrated that DBD could treat NSCLC through the synergistic correlation between multiple ingredients, multiple targets, and multiple pathways; thus, effectively improving NSCLC prognosis.

Sympathetic remodeling may cause severe arrhythmia after myocardial infarction (MI). Thus, targeting this process may be an effective strategy for clinical prevention of arrhythmias. LianXia Formula Granule (LXFG) can effectively improve the symptoms of patients with arrhythmia after MI, and modern pharmacological studies have shown that Coptidis Rhizoma and Rhizoma Pinelliae Preparata, the components of LXFG, have antiarrhythmia effects. Here, we investigated whether LXFG can mitigate sympathetic remodeling and suppress arrhythmia and then elucidated its underlying mechanism of action in rats after MI. Sprague-Dawley (SD) rats that had undergone a myocardial infarction model were randomly divided into 6 groups, namely, sham, model, metoprolol, and LXFG groups, with high, medium, and low dosages. We exposed the animals to 30 days of treatment and then evaluated incidence of arrhythmia and arrhythmia scores in vivo using programmed electrical stimulation. Moreover, we determined plasma catecholamines contents via enzyme-linked immunosorbent assay and detected expression of tyrosine hydroxylase (TH) at infarcted border zones via western blot, real-time PCR, and immunohistochemical analyses to assess sympathetic remodeling. Finally, we measured key molecules involved in the NGF/TrKA/PI3K/AKT pathways via western blot and real-time PCR. Compared with the model group, treatment with high dose of LXFG suppressed arrhythmia incidence and arrhythmia scores. In addition, all the LXFG groups significantly decreased protein and mRNA levels of TH, improved the average optical density of TH-positive nerve fibers, and reduced the levels of plasma catecholamines relative to the model group. Meanwhile, expression analysis revealed that key molecules in the NGF/TrKA/PI3K/AKT pathways were downregulated in the LXFG group when compared with model group. Overall, these findings indicate that LXFG suppresses arrhythmia and attenuates sympathetic remodeling in rats after MI. The mechanism is probably regulated by suppression of the NGF/TrKA/PI3K/AKT signaling pathway.
