The onset of puberty in mammals involves an increase in the pulsatile release of GNRH and LH. The KISS1 gene is essential for pubertal development, and its product, kisspeptin, stimulates the release of LH. The objective of this study was to determine the effects of kisspeptin in the hypothalamic-adenohypophyseal-gonadal axis of prepubertal ewe lambs. Ewe lambs (28 weeks of age) were treated intravenously with saline (control, nZ6) or kisspeptin (20 mg kisspeptin; nZ6) every hour for 24 h. Kisspeptin stimulated pulse-like release of LH within 15 min following injections, and increased the frequency and amplitude of LH pulses, and mean circulating concentrations of LH and estradiol. A surge-like release of LH was observed in four kisspeptin-treated lambs beginning 17 h after the onset of treatment, and all four lambs had elevated circulating concentrations of progesterone within 5 days post-treatment. However, circulating concentrations of progesterone decreased within 2 days after the initial rise in three of the four ewe lambs, indicating that induced luteal activity was of short duration. The proportion of lambs that were pubertal (defined by circulating concentrations of progesterone above 1 ng/ml for at least 7 days) by 35 weeks of age (8/11) and the mean age at puberty (32G1 weeks) for those reaching puberty within the experimental period did not differ between treatments. Results support a role for kisspeptin in the activation of the hypothalamic-adenohypophyseal axis leading to the onset of puberty in ewe lambs.

Abstract. This study aimed to examine the response of luteinizing hormone (LH) secretion and ovarian steroid profile to TAK-683, an investigational metastin/kisspeptin analog, through treatment during different stages of the luteal phase in goats. Nine cycling Shiba goats (4.4 ± 2.3 years old) were assigned to early luteal phase (ELP, n = 4), mid-luteal phase (MLP, n = 4), and control (n = 5) groups. The ELP and MLP groups were administered 50 µg of TAK-683 intravenously on either day 5 or between days 7-14 after ovulation, respectively. The control group received vehicle between days 7-14 after ovulation. Blood samples were collected at 10-min (2-6 h), 2-h (6-24 h), and 24-h (24-96 h) intervals after treatment. Significant increases in plasma LH concentration were detected during the periods of 3 to 5 h and 2 to 5 h in the ELP and MLP groups, respectively. Estradiol concentrations continuously increased with the rise of basal LH secretion after TAK-683 treatment in two goats of the ELP group with a surge-like release of LH, but not in the goats without LH surge, i.e. the MLP and control group ones. Plasma progesterone concentration and the lengths of estrous cycle in all groups did not change significantly from the time before and after treatment. Present findings indicate that the responses of LH and ovarian steroids to treatment with TAK-683 depend on the stage of the luteal phase of the estrous cycle. We suggest that the stimulatory effects of TAK-683 on LH secretion are reduced in the process leading to the mid-luteal phase in cycling goats. Kisspeptin-54 or kisspeptin-10, either the full length or the C-terminal amidated 10-amino-acid sequence of kisspeptin, potently stimulates luteinizing hormone (LH) secretion in both male and female mammals [1][2][3][4][5][6][7]. Our previous study showed that the stimulatory action of kisspeptin-10 on LH secretion is attributable to the stimulation of gonadotropin-releasing hormone (GnRH) neurosecretion into the hypophyseal portal circulation in castrated goats [8]. Kisspeptin is also involved in the hypothalamic regulation of GnRH secretion by gonadal steroids. Several lines of evidence suggest that the hypothalamic kisspeptin neurons expressing receptors for androgens, estrogens, and progesterone are implicated in the negative feedback control of GnRH/gonadotropin secretion [1]. Se evaluó el efecto de la kisspeptina-10 (Kiss-10), administrada repetidamente, sobre la liberación de LH y la actividad ovárica en becerras pre-púberes. Asimismo, se determinó la asociación de IGF-1 y leptina séricas con dichos efectos. Catorce (14) becerras Holstein o Suizo Pardo por Cebú (11.2±0.2 meses de edad; 187.4±6.3 kg de peso), se asignaron aleatoriamente para recibir cada dos horas, durante tres días una aplicación i.v. de Kiss-10 (Kisspeptina- 10; 5 μg/kg de peso; n=7) o solución salina fisiológica (Testigo, n=7). Se colectó sangre cada 15 min de las 0-6, 24-30 y 72-78 h, y cada 2 h hasta las 84 h. Cada aplicación de Kiss-10 fue seguida de un incremento de LH, cuya magnitud (áreabajo la curva) disminuyó (P<0.05) con el tiempo. La Kiss-10 indujo una oleada preovulatoria de LH y formación de cuerpo lúteo en 28.5 % de las becerras, mismas que presentaron las mayores (P<0.05) concentraciones séricas de IGF-1 y las menores (P<0.05) de leptina. Durante 63 días postratamiento no se detectaron cambios en las ondas foliculares atribuibles a Kiss-10 (P>0.05), y ninguna becerra manifestó actividad ovárica cíclica. Se concluye que la Kiss-10 administrada repetidamente a becerras pre-púberes es un estímulo suficiente para evocar incrementosconsistentes de LH, para inducir la ovulación y actividad lútea en animales con concentraciones séricas elevadas de IGF-1 y bajas de leptina, pero no para inducir la ciclicidad estral.
