Insights into the interaction of methotrexate and human serum albumin: A spectroscopic and molecular modeling approach

Abstract: In this study, fluorescence spectroscopy and molecular modeling approaches were employed to investigate the binding of methotrexate to human serum albumin (HSA) under physiological conditions. From the mechanism, it was demonstrated that fluorescence quenching of HSA by methotrexate results from the formation of a methotrexate/HSA complex. Binding parameters calculated using the Stern-Volmer method and the Scatchard method showed that methotrexate binds to HSA with binding affinities in the order 10 L·mol . Th… Show more

“…The full fluorescence quenching of MTX was observed at very low concentrations (less than 0.5 mg/mL), demonstrating the strong binding of MTX with plasma protein, as reported previously [78]. Taking into account the strong interaction of MTX and almost inert behavior of LDH, the fluorescence quenching of MTX-LDH could be interpreted as the action of surface-adsorbed MTX on LDH particles [79,80]. To determine the type of quenching occurring in MTX-LDH, the experimental data were fitted to several models, including the Stern-Volmer, Perrin, and polynomial equations [81], as shown in Figure S6 and Table 1.…”

“…Hydrophobic interactions also contributed to the binding between BSA and MTX, evidenced by the entropically favorable isotherms (ΔS ∼ 18 cal/mol °C at 25 °C and ionic strength of 15 mM). 63 Collectively, we determined electrostatic attractions as the main determinant to the binding between BSA and MTX, which can be modulated by changing the ionic strength of the solution.…”
