Injectable perlecan domain 1-hyaluronan microgels potentiate the cartilage repair effect of BMP2 in a murine model of early osteoarthritis

Abstract: The goal of this study was to use bioengineered injectable microgels to enhance the action of bone morphogenetic protein 2 (BMP2) and stimulate cartilage matrix repair in a reversible animal model of osteoarthritis (OA). A module of perlecan (PlnD1) bearing heparan sulfate (HS) chains was covalently immobilized to hyaluronic acid (HA) microgels for the controlled release of BMP2 in vivo. Articular cartilage damage was induced in mice using a reversible model of experimental OA and was treated by intra-articula… Show more

“…MSCs have been induced to differentiate along the chondrogenic pathway using BMPs (61,64). BMP2 has been demonstrated as among the most chondrogenic growth factors, promoting chondrocyte differentiation in vitro and augmenting articular cartilage healing in vivo (6,30,40,50,52). Bone marrow-derived mesenchymal stem cells were shown to produce hyaline-like cartilage tissue (66).…”

“…Many HA-derived medical products have been developed and tested and the readers are referred to recent reviews [17, 175] for in-depth analyses. In this article, we highlight our own experiences in the application of HA-based hydrogels for the repair and regeneration of healthy functional tissues, such as vocal folds [176179], cartilage [180, 181] and salivary glands [182, 183], as well as the creation of pathological tissue models, such as tumor spheroids [184186]. …”

“…In this instance, perlecan is likely to appear both to re-establish tissue boundaries, and also to provide a depot for the timed delivery of growth factors needed for new bone formation. Additionally, perlecan domain I has been used as a vehicle for delivery of BMP2 for healing of early lesions in an induced model of osteoarthritis or in bone repair (Decarlo et al, 2012; Srinivasan et al, 2012). …”
