Injectable and Self-Healing Hydrogel Based on Chitosan-Tannic Acid and Oxidized Hyaluronic Acid for Wound Healing

Abstract: Self-healing performance plays an important role in the in situ microinvasive injection of hydrogels, which can reduce sudden drug release and prolong the service life of hydrogels. In this paper, a multifunctional injectable and self-healing hydrogel for wound healing was developed. Chitosan (CS) was modified with TA to achieve potential adhesion, anti-inflammatory properties, and slower degradation rate. The hydrogel was formed by Schiff base reaction based on amino groups in CS and aldehyde groups in oxidiz… Show more

“…The Schiff reaction is a chemical reaction involving a dy-namic covalent imine bond formation via the crosslinking of amine groups and aldehyde groups [ 112 ]. Taking into consideration that human tissues have a large number of amino groups, the aldehydes or ketones reaction further facilitates the binding of hydrogels with biological tissues.…”

“…Therefore, this new injectable and aerosolized hydrogel has good application prospects in the fields of hemostasis and wound dressing. Similarly, by reacting TA-modified CS with oxidized hyaluronic acid (OHA), Liu et al [ 112 ] prepared a composite CS hydrogel with hemostatic, adhesive, anti-inflammatory, and slow degradation properties. The CS/TA-OHA hydrogel exhibited good self-healing properties due to the presence of dynamic reversible covalent bonds and reversible hydrogen bonds.…”

“…TA is easily oxidized (especially when pH > 7), and the phenolic hydroxyl group of the TA can form reversible covalent Schiff base (CN) interactions with amines after being oxidized to quinone. 31 Therefore, the obtained N 1s energy spectra before and after LbL assembly were deconvolved to verify whether covalent bonds were generated. Since all the N elements are from Lyz, the variation in the composition of the N peaks in Lyz and (TA-Lyz) 6-pH coatings was detected.…”

“…HA has been found to be a promising candidate to promote wound healing by facilitating the inflammatory response, angiogenesis, and tissue regeneration processes. 13,14 Collagen is a key component of the skin's ECM and is essential for dermal collagen fibrillogenesis and tissue integrity. 15,16 However, collagen has some limitations in tissue engineering applications, including its immunogenicity and low efficiency.…”
