Initiation and culture of potato tuber callus tissue with picloram

Abstract: Callus cultures of 7 potato cultivars were initiated from tuber tissue and maintained on Gelrite-solidified media with l-20pM picloram as the only PGR. Ten PM picloram was the optimal concentration for callus induction. By 4-6 weeks after explanting, there was suthcient callus produced for subculture to maintenance media which contained l-20pM picloram as the only PGR. When grown in the dark at 25'C, subcultured callus typically increased IO-fold in wet weight in 4-5 weeks. The callus produced was friable and … Show more

“…The present finding that both leaf and internodal explants of potato (cv. Iwa) exhibited a high frequency of callus formation in response to picloram alone was similar to that in a previous study on Lemhi Russet potato (Hagen et al 1990). In addition, the finding that both leaf and intermodal explants could also form callus in response to BA in combination with NAA was similar to that of a previous study on leaf sections of potato cv.…”

“…Many studies have shown that callus induction in potato could be influenced by a variety of factors in- cluding genotype, explant type, and plant growth regulator (type and concentration) (Hagen et al 1990;JayaSree et al 2001;Yee et al 2001;Tican et al 2008;Dokhaniyeh et al 2011;Pal et al 2012). The present finding that both leaf and internodal explants of potato (cv.…”

“…Picloram is one of the potent synthetic auxin useful for the initiation and maintenance of callus, suspension cultures and induction of somatic embryos from various plant tissues (Valverde et al 1987;Hagen et al 1990). The first flower appeared after 2 weeks of transfer of plants on the new medium and ten to fifteen flowers per explant were developed within the next 2 weeks.…”

“…Picloram is a potent auxin useful for the initiation and maintenance of callus and suspension cultures from certain plant tissues (Kefford & Caso 1966;Huang & Chi 1988;Hagen et al 1990). While the stability of some auxins during cell-free incubation has been investigated, there have been no similar tests with picloram.…”
