Inhibitory TCR Coreceptor PD-1 Is a Sensitive Indicator of Low-Level Replication of SIV and HIV-1

Abstract: Ongoing antigenic stimulation appears to be an important prerequisite for the persistent expression of programmed death 1 (PD-1), an inhibitory TCR coreceptor of the CD28 family. Although recent publications have emphasized the utility of PD-1 as a marker for dysfunctional T cells in chronic viral infections, its dependence on antigenic stimulation potentially renders it a sensitive indicator of low-level viral replication. To explore the antigenic threshold for the maintenance of PD-1 expression on virus-spec… Show more

“…Real-time qPCR assays that collectively or specifically detect and measure sequences associated with identification of the respective viruses in each challenge combination were applied to determine each respective viral load. Assays for total virus, SIVmac239/251 complete nef versus SIVmac239⌬nef, and SIVsmmE543/E660 versus SIVmac239/251 (gag based) were performed as previously described (13)(14)(15).…”

In our patient cohort, the frequency of activated CD4 ϩ and CD8 ϩ PD-1 ϩ CD38 ϩ cells was very significantly increased in rectal mucosae of noncontrollers compared to controllers (P ϭ 0.002 and P ϭ 0.001) or seronegative subjects (P ϭ 0.0002 and P ϭ 0.0002) (Fig.…”
