Inhibitory Effect of Magnolol and Honokiol fromMagnolia obovataon Human Fibrosarcoma HT-1080 Invasivenessin vitro

Abstract: We investigated the inhibitory effect of Magnolia obovata Thunb. bark ethanol extracts on human fibrosarcoma HT-1080 cells invasion in a reconstituted basement membrane [Matrigel (MG)]. We found that the effective components of the bark ethanol extracts were magnolol and honokiol, two biphenyl compounds. The extracts, magnolol and honokiol, did not affect HT-1080 cells adhesion to MG, but did inhibit HT-1080 cells migration at a high concentration (100 microM). HT-1080 cells secrete matrix metalloproteinase (M… Show more

“…The results demonstrated that honokiol had a similar effect to adriamycin in prolongation of lifespan of tumor-bearing nude mice. Previous studies suggest that growth inhibition by honokiol resulted from the induction of apoptosis in several cell lines [7][8][9][10][11] . A further study reported that in human squamous lung cancer cells, honokiol induced apoptosis by down-regulating Bcl-X L and sequentially activating Caspase cascade [10] .…”

“…With its major active constituent extracted from the bark of Houpu, honokiol has been found having a variety of pharmacological effects, such as anti-inflammatory [2] , antithrombotic [3] , anti-arrhythmic [4] , antioxidative [5] and anxiolytic effects [6] . Recently, honokiol has been reported to exhibit a potent cytotoxicity by inducing cell apoptosis in rat and human leukemia cells [7,8] , human fibrosarcoma cells [9] , human squamous lung cancer CH27 cells [10] and human SVR angiosarcoma cells [11] , yet there has been no report on honokiol in the treatment of human colorectal carcinoma.…”

“…Recent studies have provided experimental evidence to document anticancer effects of honokiol (5 -14). For example, the ethanol extract of Magnolia obovata and honokiol inhibited the migration of HT-1080 human fibrosarcoma cells (6). Honokiol treatment caused apoptotic cell death in CH27 human squamous lung cancer cell line in association with the up-regulation of Bad and down-regulation of Bcl-xL protein expression, release of cytochrome c from the mitochondria to the cytosol, and activation of caspases (7).…”

“…Previous studies suggest that growth inhibition by HNK resulted from the induction of apoptosis in several cell lines [9][10][27][28][29]. A further study reported that HNK induces a necrotic cell death through the mitochondrial permeability transition pore [30].…”
