Background and Aim: Drug-induced nephrotoxicity is a renal dyfunction that arises as a result of exposure to nephrotoxic drugs. Anti-tuberculosis therapy can cause nephrotoxicity and permanent kidney damage. The current study was designed to evaluate the possible protective effects of Ruta graveolens L. leaves extract against isoniazid/rifampicin-induced nephrotoxicity in rats. Methods:The experimental rats received isoniazid and rifampicin at dose level of 50 mg/kg, and 50 or 100 mg/kg/day Ruta graveolens leaves extract orally for 45 days.Results: Isoniazid/rifampicin administration induced kidney injury evidenced by the histopathological alterations as well as significant (P<0.001) increase in serum creatinine, urea and uric acid. Isoniazid/rifampicin-intoxicated rats exhibited a significant increase in serum tumor necrosis factor alpha (P<0.001), and renal lipid peroxidation (P<0.01) and nitric oxide (P<0.001) levels. On the other hand, reduced glutathione level, and activity of superoxide dismutase and glutathione peroxidase were markedly (P<0.001) declined in kidney of isoniazid/rifampicin-induced rats. Concomitant supplementation with either 50 or 100 mg/kg dose of Ruta graveolens leaves extract prevented the isoniazid/rifampicin-induced biochemical and histopathological alterations. Conclusion:The present investigation confers new information on the protective mechanism of Ruta graveolens leaves extract on anti-tuberculosis therapy-induced nephrotoxicity. Ruta graveolens protects against isoniazid/rifampicin-induced renal injury through attenuation of inflammation and oxidative stress, and potentiation of the antioxidant defense system.

Objective: The current study determines the protective efficacy of the brown seaweeds Turbinaria ornata (Turner) J. Agardh (Sargassaceae) and Padina pavonia (Linnaeus) J.V. Lamouroux (Dictyotaceae) against liver injury induced by azoxymethane (AOM). Materials and methods: Male Swiss mice received 10 mg/kg AOM once a week for two consecutive weeks and then 100 mg/kg daily dose of either T. ornata or P. pavonia ethanolic extract. Thirteen weeks after the first AOM administration and 24 h after the last treatment, overnight fasted mice were sacrificed and samples collected. Results: Compared with the AOM group, both T. ornata and P. pavonia significantly decreased the activity of aminotransferases and the concentration of bilirubin while increased albumin levels in the serum. The antioxidative effect of both extracts was observed from the increased activity of superoxide dismutase and glutathione peroxidase activities in the liver, both of which were decreased by AOM. Moreover, the levels of malondialdehyde and nitric oxide were reduced, and histological findings also confirmed the antihepatotoxic activity. In addition, treatment with T. ornata and P. pavonia significantly increased PPARc and decreased NF-jB expression in the liver of AOM-administered mice. Discussion and conclusion: Our findings indicate that the protective function of T. ornata and P. pavonia on AOM-induced liver injury may be possibly exerted by multiple pathways including abolishment of inflammation and oxidative damage, and activation of PPARc. Cyclophosphamide (CYP) belongs to the anticancer drugs that exhibit potentially life-threatening effects on the immune and hematopoietic systems. The study aimed to assess efficiency of carob pods extract against the adverse CYP effect on spleen and lymphatic tissues which are the most sensitive components of immune system. In the present study, the hematological indices and markers of oxidative stress were assessed. In addition, the native protein, lipoprotein patterns, calcium moiety of native protein, catalase (CAT), peroxidase (GPx), α-and β-esterase (EST) isoenzymes and genomic DNA patterns were electrophoretically detected in both of bone marrow and spleen tissues. It was verified that CYP caused significant (P<0.05) decline in the hematological measurements which restored in all carob treated groups. Furthermore, all native electrophoretic patterns and isoenzymes in the bone marrow and spleen tissues illustrated that the lowest similarity index (SI %) was noticed in the CYP-treated group. The carob extract increased the SI % through restoring the absent normal bands and hiding the abnormal ones in all carob treated groups. Moreover, CYP induced cleavage of the genomic DNA and the carob extract maintained integrity of the genomic DNA in spleen tissue.
