Inhibitory Action of Cyclic Guanosine 5′-Phosphoric Acid (GMP) on Oocyte Maturation: Dependence on an Intact Cumulus

Abstract: Oocyte cumulus complexes (OCC) were isolated from antral follicles of proestrous hamsters prior to the luteinizing hormone (LH)/follicle-stimulating hormone (FSH) surge and then incubated from 4 to 24 h at 37 degrees C. Under these conditions approximately 89% of the oocytes exhibited metaphase plates (11% were in the dictyate stage) and addition of LH (200 ng/ml ovine NIH-LH-S20) did not significantly alter the percentage of oocyte maturation. Approximately 50% of the OCC incubated for 24 h with 6 mM 8-bromo-… Show more

“…Recent observations have re-proposed a role for cGMP in the control of oocyte meiotic arrest and meiotic maturation [21], [22], [41], [42], [43]. LH causes a profound decrease in cGMP in the follicle and in the oocyte, which in turn should lead to activation of PDE3A.…”

“…A functional role of cGMP in inhibition of spontaneous oocyte maturation has been postulated [34]. Subsequently, Tornell et al [35] demonstrated that ANP-stimulated accumulation of cGMP in isolated rat oocyte-cumulus cellular complexes led to inhibition of spontaneous oocyte maturation.…”

“…Because cGMP can delay spontaneous maturation in rat oocytes [27] and the cAMP:cGMP ratio increases before oocyte maturation [28], we explored possible roles of cGMP by using dbcGMP and the fairly specific inhibitor of PDE5, zaprinast, which is a PDE5 antagonist in somatic cells [29]. When dbcGMP was added to HxϩFF-MAS-cultured oocytes, %GVBD was significantly lower than in the HxϩFF-MAS control group (21.9% Ϯ 8.0% vs. 82.4% Ϯ 5.1%) ( Table 1).…”
