Inhibition of the Renin-Angiotensin System Reduces Gene Expression of Inflammatory Mediators in Adipose Tissue Independent of Energy Balance

Abstract: Obesity is a growing health problem worldwide. The renin-angiotensin system (RAS) is present in adipose tissue, and evidence suggests that it is involved in both diet-induced obesity and the inflammation associated with obesity. The present experiments determined the effect of (1) different angiotensin-converting enzyme (ACE) inhibitors (captopril, perindopril, enalapril) and angiotensin receptor blockers (ARBs: telmisartan, losartan) on adiposity of mice fed a high-fat diet for 28 days (2); acute treatment wi… Show more

“…In obesity, adipocytes suffer hypertrophic and hyperplasic growth and are under stress [30,53]. The RAS is present in adipose tissue [54] and the production of RAS components by adipocytes is dysregulated by obesity, contributing to dysregulation of the systemic RAS and its consequences such as hypertension, insulin resistance in diabetes, inflammation, and oxidative stress [55]. Consistent with this, the treatment of MetS patients with the AT1 blocker irbesartan induced a significant reduction of blood pressure together with a reduction in cardiovascular risk factors: HDL cholesterol, serum triglycerides, fasting blood glucose, and waist circumference [56].…”

“…Our research supports the results of other studies on the reduction of TNFα by ACEI. Captopril used in coronary artery disease and ischemic heart diseases reduced inflammation in vivo by lowering the level of IL-6 [ 11 , 12 , 18 , 19 , 22 , 34 , 35 , 36 , 37 , 38 ], and increasing the level of TGFβ since the macrophages are in vivo activated with drugs [ 37 , 38 , 39 ]. Other authors found that non-stimulated macrophages can express the undetectable concentration of TGFβ in cell culture supernatant [ 40 ].…”

“…In the present study, body composition and markers of oxidative stress, endothelial function, and atherosclerosis were not different between the aliskiren and amlodipine treatment groups. Some studies have shown that RAS inhibitors may reduce body weight [13] and VFA [14]. Aliskiren has been reported to have an anti-inflammatory effect [30] and antioxidant activity [31].…”

“…RAS has been thought to affect body composition via its influence on the differentiation of pre-adipocytes to mature adipocytes; promotion of transcription of lipogenic enzymes, resulting in increased fatty acid synthesis and triglyceride storage; inhibition of lipolysis; and inhibition of adiponectin5 through angiotensin II type 1 receptor activation [12]. Indeed, ACE inhibitors and ARBs have been reported to decrease body weight in mice [13]. In addition, a few clinical studies have shown a decrease in visceral fat area (VFA) by ARBs [14].…”
