Inhibition of the DNA-Dependent Protein Kinase Catalytic Subunit Radiosensitizes Malignant Glioma Cells by Inducing Autophagy

Abstract: DNA-dependent protein kinase (DNA-PK) plays a major role in the repair of DNA double-strand breaks induced by ionizing radiation (IR). Lack of DNA-PK causes defective DNA doublestrand break repair and radiosensitization. In general, the cell death induced by IR is considered to be apoptotic. On the other hand, nonapoptotic cell death, autophagy, has recently attracted attention as a novel response of cancer cells to chemotherapy and IR. Autophagy is a protein degradation system characterized by a prominent for… Show more

“…42 With regard to PRKDC, its connection to the autophagy pathway is less clear, albeit there is evidence suggesting the presence of such a link. 38,43,44 The mobilization of autophagy via these DDR transducers can be prosurvival 38,39,[45][46][47][48] or antisurvival; 38,39,48,49 and the dual roles that autophagy plays in cell fate determination under DNA damage stress conditions add to the complexity of its involvement in DDR, and may influence the outcome of anticancer chemotherapy. The interplay between DDR and autophagy is a burgeoning area of research in the autophagy field, linking autophagy with genomic integrity.…”

“…To detect and quantify acidic vesicular organelles, cells were stained with acridine orange as described previously (38). The number of acridine orange -positive cells was determined by fluorescence-activated cell sorting (FACS) analysis.…”

“…Plated cells were transfected with double-stranded siRNA targeting TG2 mRNA (Qiagen, Venlo, the Netherlands), PKCy (Santa Cruz Biotechnology, Santa Cruz, CA), and Beclin-1 (Santa Cruz Biotechnology) and control (nonsilencing) siRNA (Qiagen) and/or the GFP-LC3 fusion vector (22,38) using the Qiagen transfection reagent according to the manufacturer's protocol. Two siRNA sequences targeting TG2 were designed using siRNA-designing software (Qiagen).…”

“…GFP-LC3 cells present a diffuse distribution under control conditions, whereas a punctate pattern of GFP-LC3 expression (GFP-LC3 dots) is induced by autophagy. In the current study, we determined the prevalence of autophagic cells (i.e., cells expressing GFP-LC3 dots) by using the GFP-LC3 expression vector kindly provided by Dr Noboru Mizushima (Tokyo Metropolitan Institute of Medical Science, Tokyo, Japan), as described previously (Daido et al, 2005;Kanzawa et al, 2005). Tumor cells were transiently transfected with this vector by using Fugene 6 transfection reagent (Roche).…”
