Inhibition of the Cutaneous Response to Histamine by H1-Blocking Agents

Abstract: A controlled, randomized, double-blind, cross-over study was performed in 10 healthy volunteers in order to compare changes of cutaneous blood flow values (CBFV, laser Doppler flowmetry, Periflux, Perimed, S) in histamine-challenged skin before and after administration of a capsule containing either 10 mg cetirizine or 60 mg terfenadine. The overall pattern of the basal cutaneous wheal and flare response to the agonists (histamine: aqueous solution: Hsol, or Phazet®: Hprick) and control saline solution, showed… Show more

“…As there is more and more liquid in the interstitial dermal tissue, the wheal appears and the skin volume between vessels increases along with the increased dermal pressure, reflecting the elastic properties of the skin. As a result, LDM data reach lower levels than the stable high levels measured in the flare area, confirming previous observations 10,11,21–23 …”

“…This may lead to aberrant results (19, 20) (i.e., inhibitions greater than 100% or equivalence between different doses), in total contradiction of the literature. On the contrary, the injection of a small volume (10 µl) of saline induces very small perturbation, as shown by van Neste & Rihoux (21). We therefore believe that the injection of a small volume is more specific from a pharmacologic point of view.…”
