Inhibition of the Calcium Release-activated Calcium (CRAC) Current in Jurkat T Cells by the HIV-1 Envelope Protein gp160

Abstract: The HIV-1 envelope glycoprotein gp120/160 has pleiotropic effects on T cell function. We investigated whether Ca 2؉ signaling, a crucial step for T cell activation, was altered by prolonged exposure of Jurkat T cells to gp160.

“…Depending on the specifically tested cells and the experimental conditions, variable results have been reported on the modulation of SOC by PKC (1,2,7,39,46,51). The differential tissue distribution, intracellular localization, and cellular functions of different isoforms of PKC might also contribute to these discrepancies.…”

“…L15 fibroblasts stably overexpressing IP 3 R1 [35] were a generous gift of Pr Mikoshiba (RIKEN Brain Science Institute, Japan). They were maintained in DMEM medium (Gibco®, Life Technologies, Gent, Belgium) supplemented with 10 % heat-inactivated fetal calf serum, 3.8 mM Lglutamine, 1% non-essential amino acids, 85 IU ml -1 penicillin, 85 g ml -1 streptomycin, 400 ] cyt subsequently to Ca 2+ calcium influx [37]. At the end of each experiment, 1 µM GdCl 3 was added to verify that MDEB only acts on the SOCE [17].…”

“…I CRAC is the plasma membrane Ca 2+ current activated by passive depletion of the internal stores [37]. Upon break-in with the pipette solution containing 10 mM BAPTA, hyperpolarization of the Jurkat cell membrane evoked a weak inward current in the absence of external Ca 2+ ions (Fig.…”
